体外循环再灌注血清对人肾小管上皮细胞的损伤作用

         

摘要

Objective To investigate the effect of the serum after cardiopulmonary bypass (CPB) on human proximal tubular epithelial cells (HK-2) in vitro by improved ischemia reperfusion model. Methods HK-2 cell was used as target cell. The experiment was divided into 3 groups by culture medium, blank group ( human serum-free medium) , control group ( medium containing normal human serum) , test group ( medium containing CPB serum, hypoxia and reoxygenation cultured). The apoptosis rate of HK-2 cell was detected by flow cytometry. Caspase-3 protein expression was detected by immunohistochemical technique. Result The apoptosis rate of HK-2 cell and Caspase-3 in test group were significantly higher than blank group and control group (P <0. 05) , but there was no significant difference between blank group and control group. Conclusion CPB serum can induce HK-2 cell apoptosis, which mechanism may be related to the activation of cellular Caspase-3.%目的 通过改良缺血再灌注模型,观察体外循环血清对人肾小管上皮细胞损伤的作用.方法 以人近曲肾小管上皮细胞(HK-2)为研究对象,根据是否应用体外循环血清将细胞分为空白组:不含人血清的培养基;对照组:含正常人血清的培养基;实验组:含体外循环血清的培养基,进行缺氧复氧培养.通过流式细胞仪检测肾小管细胞凋亡情况,免疫组化检测Caspase-3蛋白表达.结果 实验组凋亡率及Caspase-3均高于对照组及空白组(P<0.05);空白组和对照组凋亡率及Caspase-3表达差异无统计学意义.结论 体外循环血清可诱导HK-2细胞凋亡,其机制可能与激活胞内Caspase-3有关.
