Objective To investigate the influence and underlying mechanisms of hydrogen peroxide (H2O2) on the survival promoting effect of insulin-like growth factor 1 (IGF-1) in PC12 cells. Methods Pretrearment with different concentrations of H,0, in PC12 cells for 60 minutes, MTT assay and Western blot were performed to examine H2O2 in influencing the survival promoting effect of IGF-1 and the tyrosine phosphorylation of IGF-1 receptor. We investigated the underlying mechanisms by using different antagonists. Results Pretrearment of H2O2 could attenuate survival promoting effect of IGF-1 and its receptor tyrosine phosphorylation in a dose-dependent manner, while MK801 could reserve the inhibitory effect of H2O2, Conclusion H2O2 inhibits IGF-1 receptor tyrosine phosphorylation and the survival promoting effect by NMDA receptor.%目的 研究过氧化氢( H2 O2)对PC12细胞胰岛素样生长因子(IGF-1)促存活功能的影响及可能机制.方法 不同浓度的H2O2预处理PC12细胞60 min,MTT及Western blot方法观察H2 O2对IGF-1促存活及其受体酪氨酸磷酸化的影响;不同的抑制剂预处理研究H2O:影响IGF-1促存活功能的可能机制.结果 H2O2预处理可以剂量依赖性的降低PC12细胞IGF-1的促存活功能及其受体的磷酸化水平,NMDA受体的特异性拮抗剂MK801可以逆转H2O2的抑制作用.结论 H2O2可以通过谷氨酸受体亚型NMDA受体抑制IGF-1R的磷酸化,从而抑制其促存活作用.
