羧氨基葡聚多糖钠对胃癌脱落细胞种植转移的抑制作用

摘要

目的：探讨羧氨基葡聚多糖钠抑制胃癌脱落细胞种植转移的效果。方法取人胃癌MGC-803细胞株，分别加入12．5、25、50 mol/L羟氨基葡聚多糖钠，另设对照组加入生理盐水。采用MTT法检测胃癌细胞增殖情况，流式细胞仪检测细胞周期和凋亡情况，Transwell小室检测细胞迁移能力。结果羧氨基葡聚多糖钠显著抑制胃癌细胞的增殖，并呈剂量依赖性抑制（P＜0．01）；羧氨基葡聚多糖钠可引起胃癌细胞周期G1/S期阻滞和细胞凋亡，显著抑制胃癌细胞的转移（P均＜0．01）。结论羧氨基葡聚多糖钠具有显著的抑制胃癌细胞生长及转移效应，可作为进展期胃癌根治性手术后腹腔冲洗液用于临床。%Objective To investigate the effect of Surg take on cell growth and migration in Gastric cancer cell MGC-803 .Methods Cell proliferation of Gastric cancer cell with different concentration of Surg take solution was examined by MTT assay;Cell cycle distribution and cell apoptosis was determined by flow cytomtry .Cell migration assay was assessed by Transwell chambers .Results Surg take solution could suppress the growth of MGC-803 cells in gastric carcinoma , in a dose-dependent manner (P<0.01).Surg take solution could induce cell cycle arrest at the G 1/S phase and cellular apop-tosis, and Surg take solution could significantly suppress the migration of MGC-803 cells in gastric carcinoma ( all P <0.01).Conclusion Surg take could suppress the growth and migration of gastric carcinoma cells , which may provide a prominent choice for irrigation of peritoneal cavity for the radical operation of advanced gastric carcinoma .
