一株鹅源 H9 N2亚型流感病毒的分离鉴定及其 HA 基因的遗传进化分析

         

摘要

One avian influenza virus strain named A/goose/Shandong/XJ/2014 ( H9 N2 ) was isolated from goose in Shandong in 2014, and it was confirmed by HI cross-inhibition test and gene cloning.The HA and NA gene of the isolated strain were cloned and sequenced to analyze their molecular characterization and hereditary variation.The deduced amino acid sequence of HA gene showed that it shared a lentogenic motif PSRSSRGLF at the cleavage site.The nucleotide sequence homologies of HA gene of XJ with that of the refer-ence strains were 87.2%~99.9%, and the amino acid sequence homologies were 87.4%~99.8%.There was a closer genetic relationship between the XJ and H9 N2 epidemic strains in China especially those isolated after 2013 .%2014年从山东省某鹅场发病鹅体内分离到一株具有血凝性的病毒,对其进行鉴定,并对其HA基因和NA基因进行克隆测序,分析了解其与其他H9N2各毒株间的变异关系和进化规律。结果表明:经HI 交叉抑制试验和基因克隆最终证实该毒株为 H9N2亚型流感病毒,将其命名为 A/goose/Shandong/XJ/2014(H9N2),简称XJ。 XJ分离毒株的HA基因裂解位点序列为PSRSSRGLF,为弱毒毒株的特征序列。其HA基因与其他参考株HA基因的核苷酸序列同源性为87.2%~99.9%,氨基酸序列同源性为87.4%~99.8%。 XJ与国内大部分同期分离毒株遗传距离较近,尤其是2013年以后的分离毒株。
