原花青素交联去细胞牛颈静脉带瓣管道的实验研究

         

摘要

Objective: To investigate the effect of proanthocyanidin-fixed decellularized valved bovine jugular vein conduit (BJVC), and to establish the experimental foundation for the further application it in cardiovascular surgery and tissue engineering. Methods: Twenty-four BJVCs, randomly divided into 3 groups and fixed with proanthocyanidins (decellularized BJVCs fixed with 0.5% proanthocyanidin), glutaraldehyde ( decellularized BJVCs fixed with 0.625% glutaraldehyde) and the control group (fresh BJVCs), respectively. The morphologic and mechanical properties of BJVCs in the 3 groups, including thickness, diameter, moisture content, denaturation temperature, tensile strength, elongation at break, fixed index and the in vitro enzymatic degradation resistance, were detected. And the vitro cytotoxicity test and hemolysis tests were applied in the biological evaluation of the fixed BJVCs. Results: l.The cells compounds removed thoroughly, fiber stents to maintain complete; 2 There were no significant differences in its effect of these two crosslinking agents, though the crosslinking speed of proanthocyanidin was slower than glutaraldehyde; 3 There were no difference in wall thickness, diameter between proanthocyanidin group and fresh group. Though the tissue water content of proanthocyanidin fixed tissue was inferior to fresh tissue, it was still higher than glutaraldehyde fixed tissue. For fixation of biological tissue with glutaraldehyde or proanthocyanidin, denaturation temperature, tensile strength and elongation increase at break as well as in vitro enzymatic degradation resistance. There were no significant differences in denaturation temperature and the in vitro enzymatic degradation between glutaraldehyde and proanthocyanidin fixed tissues. However, the tensile strength and elongation at break of the proanthocyanidin fixed tissueswere significantly superior to that of the glutaraldehyde fixed tissues. 4 The hemolysis tests showed that hemolysis rate of fixed tissues which fixed with glutaraldehyde or proanthocyanidin, respectively, <5%, in line with the national biological materials hemolysis test standards, but the group of fixed with proanthocyanidin was significantly lower than that of glutaraldehyde. And the vitro cytotoxicity test showed that cytotoxicity of the proanthocyanidin fixed tissues was lower than that of the glutaraldehyde fixed tissues. Conclusions: Proanthocyanidin is an effective and suitable approach for crosslink BJVC without toxicity compared with glutaraldehyde , and it has a good research and application prospect in improving the physicochemical properties of BJVC.%目的:探讨原花青素(Proanthocyanidin,PA)交联去细胞牛颈静脉带瓣管道(valved bovine jugular vein conduit,BJVC)的方法和效果,为其在心血管外科和组织工程中的应用奠定实验基础.方法:新鲜BJVC 24根,随机分为原花青素交联组(PA组,n=8),戊二醛(Glutaraldehyde,GA)交联组(GA组,n=8)及新鲜对照组(Fresh组,n=8).分别检测各组牛颈静脉带瓣管道的生物物理性能.同时采用溶血实验和细胞毒性实验对PA和GA交联组分别进行生物学评价.结果:1.细胞成分去除彻底,纤维支架保存完整;2.固定指数检测显示虽然PA较GA交联速度慢,但两种交联剂交联效果无明显差异.3.生物物理性能检测显示:PA组管道厚度、管腔直径与新鲜对照组相当(P>0.05),且优于GA组(P<0.05);PA组合水量高于GA组(P<0.05);两种方法交联后热皱缩温度、断裂强度、断裂延伸率均明显增强(P<0.05),且PA组织断裂强度和断裂延伸率优于GA组(P<0.05);Fresh组浸泡8d后失重率为98%,而PA组和GA组分别为3.68%和4.31%,两组间无显著差异.4.生物学评价结果表明,PA组细胞毒性低于GA组(P<0.05);溶血实验结果表明两组溶血率均<5%,符合国家生物材料溶血试验标准,且PA组明显低于GA组.结论:PA交联的BJVC具有良好的生物物理学特性,较低细胞毒性,为PA交联BJVC的深入研究做了初步探讨.
