Objective To simultaneously determine the contents of five active components in Lonicera japoni ca Thunb by HPLC method. Methods The separation of five ingredients was achieved on a Agilent Eclipse XDB-C18 column( 250 mm ×4. 6 mm,5 μm )with gradient elution using solvents of acetonitrile and 0. 4% phosphoric acid,the flow rate was 1. 0 mL/min. The detective wave length was at 350 nm,and the inpouring volume was 10 μL. Results The recovery rate was in the range of 97. 5% ~ 100. 2% ,and the compounds showed good linearity( rS=0. 999 3 )in a detection range. Conclusion The method is simple, sensitive, accurate and reproducible, it can be used to control the quality of Lonicera japonica Thunb.%目的 建立HPLC法同时测定金银花中5种有效成分的含量.方法 采用Agilent Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.4%的磷酸水溶液进行线性梯度洗脱,流速:1.0 mL/min,检测波长:350 nm,进样量:10μL.结果 绿原酸、咖啡酸、芦丁、木犀草苷、槲皮素5个化合物在测定的范围内均表现出良好的线性关系(r≥0.999 3),回收率为97.5%~100.2%.结论 该方法操作简便,分离效果好,灵敏度高,重复性好,为较全面控制金银花药材的质量提供了一种可靠的方法.
