ABCG2及CD133在体内外不同分化胃癌中的表达水平研究

         

摘要

Objective To determine whether quantitative relationship exists between the expressions of 2 cancer stem cell markers,ABCG2 and GDI33,and differentiation degrees of gastric cancer. Methods (1)The expressions of ABGG2 and GDI33 were detected in 78 cases of human gastric adenocarcinomas by immunoflures-cence assay;(2)2 markers were also detected in 3 gastric cancer cell lines, undifferentiated gastric cancer cell line HGG - 27, poorly differentiated gastric cancer cell line; BGG - 823 , and moderately - poorly differentiated gastric adenocarcinoma cell line SGG - 7901 , in vitro by flow cytometric analysis. (3)Gastric xenotransplant cancers in nude mice were constructed to compare the malignancy of 3 separated cell lines. Expressions of 2 markers were detected in vivo by immunoflurescent staining. Results (1)Expressions of 2 markers were generally correlated with the differentiations of gastric cancers. In human gastric adenocarcinoma,the expressions of 2 markers were increased with the elevation of the malignancy grades of gastric cancers. In addition, the expressions of GD133 were higher in diffuse type than those in intestinal type of human gastric cancers. (2)Undifferentiated cell line HGG -27 expressed more markers than moderately - poorly differentiated cell line SGG - 7901. (3)The similar result had been observed in xenotransplant tumors which arouse from 3 cell lines. Conclusion The expressions of cancer stem cell markers ABGG2 and GD133 differed in various differentiation degrees of gastric cancers, poorly differentiated gastric cancer expressed relatively more.%目的 研究ABCG2及CD133两种待定肿瘤干细胞标志物的表达与不同分化胃腺癌的数量关系.方法 (1)在78例人胃腺癌组织中以免疫荧光法检测ABCG2和CD133的表达;(2)以流式细胞技术对未分化胃癌细胞系HGC-27、低分化胃癌细胞系BGC-823及中-低分化胃腺癌细胞系SGC-7901中ABCG2、CD133的表达进行测定;(3)用上述三种细胞系建立裸鼠胃壁移植成瘤模型,采用免疫荧光染色的方法对体内环境下ABCG2和CD133在移植瘤中的表达情况进行检测.结果 (1)ABCG2和CD133的表达均与肿瘤分化程度相关.在人胃腺癌标本中,两种标志物的表达随着胃癌恶性程度的增加而增加,分化越差的胃癌ABCG2、CD133表达水平越高;并且根据Lauren分型,CD133在人弥散型胃癌中的表达高于肠型胃癌;(2)未分化细胞系HGC-27中ABCG2和CD133的表达要比中-低分化细胞系高;(3)三种胃癌细胞移植鼠模型中移植瘤组织ABCG2和CD133表达水平与人胃癌组织和不同分化胃癌细胞系的表达水平相似,低分化胃癌细胞株HGC-27移植瘤ABCG2、CD133表达水平最高.结论 肿瘤干细胞标志物ABCG2和CD133在不同分化胃癌中的表达有差异,且低分化胃癌其肿瘤干细胞标志物表达水平较高.
