钙镁合剂联合谷胱甘肽减少奥沙利铂相关神经毒性的随机对照研究

             

摘要

目的 探讨钙镁合剂联合谷胱甘肽对奥沙利铂相关神经毒性的预防作用.方法 41例接受mFOLFOX6方案化疗的结直肠癌患者按照随机数字表法分为治疗组和对照组.治疗组(n =20)化疗前静脉滴注钙镁合剂及谷胱甘肽,对照组(n=21)化疗前静脉滴注安慰剂.观察并分析两组患者治疗后奥沙利铂相关神经毒性发生率及程度.结果 8周期化疗结束后,Ⅱ、Ⅲ级神经毒性发生率治疗组为25.0% (5/20),对照组为57.1%( 12/21),两组比较差异有统计学意义(x2 =4.361,P<0.05);两组Ⅰ级神经毒性发生率比较差异无统计学意义(40% vs 42.8%,P>0.05).结论 化疗前使用钙镁合剂和谷胱甘肽,能减少奥沙利铂剂量相关神经毒性的发生,可作为预防其神经毒性的措施.%Objective To investigate whether the administration of calcium/magnesium (Ca/Mg) infusion combined with Glu-tathione (GSH) infusion can reduce Oxaliplatin-related neurotoxicity. Methods Patients after FOLFOX6 chemotherapy were randomized into treatment group and control group,and received Ca/Mg infusion and Glutathione infusion(treatment group,n = 20) or placebo (control group,n=21) before the administration of Oxaliplatin. The incidence and severity of Oxaliplatin-induced neurotoxicity were observed. Results After eight cycles of chemotherapy,25. 0% (5/20) of patients in treatment group were observed grade Ⅱ - Ⅲ neurotoxicity while 57. 1% (12/21) in control group were. The incidence of neurotoxicity in grade Ⅱ - Ⅱ was significantly different between the two groups (χ2 =4. 361 ,P <0. 05). The incidence of neurotoxicity in grade I was not significantly different between the two groups (40% vs. 42. 8% ,P <0. 05). Conclusion The therapy of calcium/magnesium infusion combined with Glutathione infusion is able to reduce the incidence of Oxaliplatin-induced neurotoxicity.
