RP-HPLC法同时测定独一味地上部分中Sesamoside、Phlorigidoside C的含量

To develop an HPLC method to determine Sesamoside and Phlorigidoside C in Herb Lamiophlomis rotata. Methods: The samples were separated at 30℃ on an Ultimate XB-C18 column (4.6 mmx250 mm, 5 μm) eluted with methanol and 0.2% phosphoric acid in a gradient mode. The ratio of methanol was kept constant of 15% from 0 to 22 min and increased to 70% in 2 min, then kept constant from 24 to 30 min and decreased back to 15% in 2 min. The detection wavelength was 237 nm, and the flow rate was 1 mL·min-1. Results: Sesamoside: Y = 1.536xl03X-1.151, r=0.9999; Phlorigidoside C: Y = 1.286x 103X-4.995, r=0.9999. Conclusion: The method was rapid and precise, and can be used for controlling the quality of Herb Lamiophlomis rotata.%目的:建立同时测定独一味地上部分中Sesamoside、Phlorigidoside C的反相高效液相色谱法.方法:采用RP-HPLC法,色谱柱Ultimate XB-C18(4.6 min×250 mm,5μm)；流动相为甲醇(A相)-0.2％磷酸梯度洗脱(0～22 min A相15％；22～24 min A相15～70％；24～30 min A相70％；30～32 min A相70～15％)；流速1 mL· min-1；检测波长237 nm；柱温30℃.结果:回归方程分别为Sesamoside:Y=1.536×103X- 1.151 (r=0.9999)；Phlorigidoside C:Y=1.286×103X-4.995,(r=0.9999).结论:该方法快速、准确、重复性好,为更好地控制独一味药材的质量提供了可行的方法.
