高速逆流色谱法分离纯化长春花中长春新碱

         

摘要

Objective To utilize high-speed counter-current chromatographyf HSCCC) for separation and purification of vincristine from Calharanlhus roseus , and to establish relevant process and conditions to provide data for industrial production. Method Highspeed counter-current chromatography was used with chloroform-methanol-0. 3 mol· L-1 HC1 (4 : 3 : 2) as the solvent system. The light phase was used as the stationary phase, and the other as the mobile phase. The host speed was 800 r ? Min-1 , the flow rate of mobile phase was 2. 0 ml. ? Min ' , the wavelength of detector was 280 nm, and the temperature of circulating water was 28°C. Result The purity of vincristine purificated by HSCCC was 79. 7% (detected by HPLC). Conclusion Purification of vincristine by HSCCC has many advantages,such as large capacity, high separation efficiency,and high purity. Thus,it is suitable for industrial production of high purity products.%目的 采用高速逆流色谱(HSCCC)分离纯化长春花中长春新碱,建立相关工艺条件,为工业生产提供参考数据.方法 采用高速逆流色谱仪,氯仿-甲醇-0.3 mol·L -1 HCl(4∶3∶2)溶剂体系,上相为固定相,下相为流动相,流动相流速为2.0 mL·min -1,紫外检测波长为280 nm,主机转速为800 r·min -1,恒温循环温度为28 ℃.结果 HPLC检测HSCCC纯化后长春新碱质量分数为79.7%.结论 高速逆流色谱纯化长春花中长春新碱容量大、分离纯度和效率高,避免了有机溶剂的大量使用,适用于工业生产高纯度产品.
