不同剂量内毒素诱导瘀热互结证大鼠血液流变学指标的动态观察

     

摘要

This study was aimed to discuss the dynamic variation law of hemorheology in rat model with syndrome of blood and heat stasis induced by different doses of lipopolysaccharide (LPS) in order to provide a reference for research of the biological basis and drug intervention mechanism. A total of 100 rats were randomly divided into the normal group (n = 20) and the model group (n = 80). Then, the model groups were continuously divided into 16 groups, according to the doses of 200, 100, 50, 20 μg · kg-1 and the time points of 0.5, 3, 6, 12 h, with 5 rats per group. The four doses researches were done separately. And the normal control group was established. Except normal control group, the rats were injected with corresponding doses of LPS (2 mL·kg-1). At corresponding time points, 5 mL of blood was drawn from the abdominal aorta for the hemorheology testing. The results showed that compared with the normal control group, rats of 200 ug/kg dose group (3, 6, 12 h), 100 μg · kg-1 dose group (6, 12 h), and 50 μg·kg-1 dose group (0.5, 3, 6, 12 h), whose high, medium and low shear viscosity of whole blood, hematocrit, sedimentation equation K value, erythrocyte electrophoresis time, casson yield stress indicators were significantly low (P < 0.05 or P < 0.01); coagulation tests showed low congeal and dissolution state of the blood in rats of different doses groups of LPS. It was concluded that the state of hemorheology in rat model with syndrome of blood and heat stasis induced by different doses of LPS was low viscosity, low density and low aggregation.%目的:探讨不同剂量内毒素诱导的瘀热互结证大鼠模型血液流变学指标的动态变化规律,为研究该模型的生物学基础及药物干预机制提供参考.方法:100只大鼠随机分为模型组(n=80)和正常组(n=20),模型组又按200,100,50,20 μg·kg-1 4个剂量,每个剂量0.5,3,6,12 h 4个时间点,随机分为16组,每组5只大鼠.各剂量分次进行实验,每次实验均设正常组5只大鼠.除正常组外,其余各组大鼠均经尾静脉注射1次相应剂量的内毒素(2 mL·kg-1),大鼠自由饮水、进食.于注射内毒素后各时间点,腹主动脉取血5 mL,进行血液流变学测定.结果:与相应正常组比较,200 μg·kg-1剂量的3 h组、6 h组、12 h组,100 μg·kg-1剂量的6 h组、12 h组,50 μg·kg-1剂量的0.5 h组、3 h组、6 h组、12 h组全血高、中、低切黏度,以及红细胞压积、红细胞电泳时间、卡松屈服应力等指标明显降低(P<0.05或P<0.01);凝血功能检测显示,内毒素不同剂量组大鼠血液呈现低凝溶解状态.结论:不同剂量内毒素诱导建立的瘀热互结证大鼠模型的血液流变学指标均呈低黏、低浓、低聚、低凝状态,研究结果在一定程度上反映了内毒素诱导建立的瘀热互结证大鼠模型血液流变学特征,为该模型的建立方法和条件及药物干预机制提供参考.
