农产品质量安全快速检测技术——微波辅助水解比色法快速测定动物水解蛋白中羟脯氨酸的研究

     

摘要

A rapid determination method for 4-Hydroxyproline of hydrolyzed collagen was developed, based on microwave-assisted hydrolysis and colorimetric method.In this paper the influence effects were investigated, such as microwave-assisted hydrolysis conditions (the concentration of H2SO4, volume of H2SO4, hydrolysis time and microwave power) and color development conditions (oxidation time, oxidation temperature, color developing time).The results showed that microwave-assisted hydrolysis had the same precision and accuracy compared to traditional hydrolysis method.However, the hydrolysis time by microwave-assisted hydrolysis was reduced from 16 h to 4~8 min.The novel rapid analyzer and reagent kit for 4-Hydroxyproline designed by Jilin University - Little Swan Instruments Co., Ltd.was used to detect 4-Hydroxyproline.It was found that at the concentration range of 0.10×10-3 g/L~2.50×10-3 g/L, the absorbance and concentration of 4-Hydroxyproline has a good linear relationship, and the recovery rate between 90% and 105%.%本文研究了一种微波辅助水解比色法测定动物水解蛋白中羟脯氨酸的快速检测方法及仪器.考察了微波辅助水解(硫酸浓度、体积、微波时间、功率)及比色条件(氧化温度、时间、显色时间)对样品水解和羟脯氨酸测定的影响.结果表明:通过微波辅助水解法对动物水解蛋白水解4~8 min与传统加热水解16 h所得的羟脯氨酸含量基本相同,将样品前处理时间从16 h缩短到4~8min.采用吉大·小天鹅所研制的新型羟脯氨酸快速检测仪及其配套试剂盒进行测定,羟脯氨酸在0.10x10-3g/L~2.50 x 10~3 g/L浓度范围内,其吸光度与浓度具有良好的线性关系,加标回收率90%~105%.该方法可判断掺假奶制品中是否添加了动物水解蛋白.
