葛花、枳蓂子配伍对慢性酒精性肝损伤大鼠海马7种单胺类神经递质及代谢产物含量水平的影响

     

摘要

目的:从配伍剂量、给药周期两个角度,探讨葛花、枳蓂子配伍对慢性酒精性肝损伤大鼠海马7种单胺类神经递质及代谢产物含量的影响。方法:采用白酒灌胃的方法复制慢性酒精性肝损伤大鼠模型,将 Wistar 大鼠随机分为空白组、模型组、东宝甘泰组、葛花枳蓂子配伍组(低、中、高剂量),分别于灌胃4、8、12周后,冰上快速剥取海马组织,利用高效液相-电化学方法检测大鼠海马单胺类神经递质及其代谢产物的含量。结果:造模4周后,与模型组相比,5-羟色胺(5-HT)的含量在配伍的3个剂量组有统计学差异;去甲肾上腺素(NE)的含量在配伍中、高剂量组有统计学差异;高香草酸和肾上腺素的含量在配伍中剂量组有统计学差异。造模8周后,与模型组相比,5-HT 的含量在配伍的3个剂量组有统计学差异;5-羟吲哚乙酸(5-HIAA)的含量在配伍中、高剂量组有统计学差异;多巴胺的含量在配伍中剂量组有统计学差异。造模12周后,与模型组相比,5-HIAA 的含量在配伍低、高剂量组有统计学差异;NE 的含量在配伍中剂量组有统计学差异。结论:葛花、枳蓂子配伍对慢性酒精性肝损伤模型大鼠海马单胺类神经递质及代谢产物含量有一定的调节作用,为葛花、枳蓂子配伍防治酒精性相关疾病的研究提供了数据支撑。%Objective:To examine the effectiveness of compatibility of Flos Puerariae Lobatae and Semen Hoveniae on 7 kinds of monoamine neurotransmitters and their metabolites in hippocampal tissue of rats with chronic alcoholic liver injury from two aspects-the compatibility dosage and dosing cycle.Methods:The rats model with chronic alcoholic liver injury were induced by administrating wine.The rats randomly divided into blank control group,model group,positive control group,and compatibility groups with dose difference (low,medium and high doses).After 4,8 and 12 weeks,hippocampus were rapidly removed and frozen immediately.HPLC-ECD was used to test the expression of monoamine neurotransmitters and their metabolites of hippocampal tissue in rats.Results:After 4 weeks,in comparison to model group,the content of 5-HT in the three compatibility doses groups were changed with statistically significant.The content of NE in the medium and high doses group were changed with statistically significant.The content change of HVA and E in the medium dose group were changed with statistically significant.After 8 weeks,In comparison to model group,the content of 5-HT in the three compatibility doses groups were changed with statistically significant.The content of 5-HIAA in the medium and high doses group were changed with statistically significant;the content change of DA in the medium dose group were changed with statistically significant.After 12 weeks,In comparison to model group,the content of 5-HIAA in the low and high doses group were changed with statistically significant.The content change of NE in the medium dose group were changed with statistically significant.Conclusion:Compatibility of Flos Puerariae Lobatae and Semen Hoveniae could affect the expression of monoamine neurotransmitters and their metabolites in hippocampal tissue of rats with chronic alcoholic liver injury.
