PCR引物设计方法综述

摘要

In order to obtain an ideal method of PCR primer design, the author completed primer design of target gene, using some databases and software tools for primer design, such as NCBI, Primer Premier 5.0 and so on, combined with the principle of primer design.Then made a research on optimization of conditions for PCR reaction after the designed primer was analyzed through BLAST comparison, comprehensive evaluation and gene retest analysis. The results indicated that primers designed through these databases and softwares could basically meet the requirements of following RTPCR reaction, and could make an in-depth study of molecular biology.%为了获得PCR引物设计的理想方法．笔者运用NCBI等数据库与引物设计软件Primer Premier 5．0等工具，结合引物设计的原则，完成目的基因的引物设计，并对设计后的引物进行BLAST比对、综合评价和基因的重新检测等分析，据此对PCR反应条件进行优化研究。结果表明，通过这些数据库和软件所设计的引物能基本满足后续的RT—PCR反应，可以进行进一步的分子生物学研究。
