结核分枝杆菌抗原蛋白38×103抗原表位预测及其蛋白片段抗原性研究

     

摘要

Objective To screen a specific diagnostic marker of Mycobacterium tuberculosis(MTB). Methods The 38 x 10 epitope in MTB was analyzed and predicted , and the 121 - 182 and 238 -374 amino acid fragments of 38 x 10 in prokaryotic vectors were named T38 and P38 recombinant proteins ; their antigenicity was detected by protein chips . Results The protein chip was applied to detect 94 positive and 128 negative samples. The results showed certain sensitivity (81. 9% ) and high specificity (80. 5% ) when P38 recombinant protein was used as an antigen for tuberculosis diagnosis , but T38 had low specificity. Conclusion Bioinformatics is applied to predict the 38 x 10 epitope, so that a better target site of antigens can be fourd. P38 recombinant protein can be potentially used for the diagnosis of tuberculosis and as a can -didate antigen.%目的 筛选出一种结核分枝杆菌(MTB)特异性诊断标志物.方法 对MTB重组蛋白抗原38×103的基因组序列进行生物信息学分析及抗原表位预测,原核表达38×103基因序列121~182和238~374位氨基酸蛋白片段,分别命名为T38和P38重组蛋白;采用蛋白芯片对T38和P38重组蛋白进行抗原性检测.结果 经蛋白芯片检测94例阳性和128例阴性样本,结果显示,P38重组蛋白在结核病检测中的敏感性为81.9%,特异性为80.5%,但是T38重组蛋白特异性不佳.结论 生物信息学预测38×103基因抗原表位,为寻找有价值的抗原靶点提供了参考;重组蛋白片段P38在结核病诊断中具有潜在应用价值,可作为联合诊断的备选抗原之一.
