依达拉奉对脑梗死大鼠神经保护作用最佳治疗时间窗的探讨

             

摘要

目的:采用大鼠大脑中动脉梗塞( MCAO)模型,探讨依达拉奉治疗急性脑梗死的最佳治疗时间窗。方法将MCAO模型大鼠随机分为A、B、C、D和E组(n=10)。 A、B、C、D组分别于造模后0.5 h、1 h、1.5 h、2 h通过尾静脉给予3 mg/kg的依达拉奉,E组于造模后给予等量的生理盐水。于梗死后24 h进行神经功能缺损评分,并测量脑梗死体积。结果 A、B、C组神经功能缺损评分和脑梗死体积均低于E组(P<0.05,P<0.01),其中B组与E组比较差异更显著(P<0.01),B组低于A、C、D组(P<0.05,P<0.01),B组与D组比较差异更显著(P<0.01)。结论对大鼠MCAO模型,依达拉奉具有神经保护作用,且其神经保护作用有治疗时间窗。%Objective To investigate the golden hour of Edaravone neuroprotective effect in treatment of rats with acute cerebral infarction using middle cerebral artery occlusion ( MCAO) models. Methods A total of 50 MCAO rat models were randomly divided into group A, B, C, D and E ( n=10 in each group) . Group A, B, C and D were re-spectively injected with 3 mg/kg Edaravone via caudal vein infusion at 0. 5 h, 1 h, 1. 5 h and 2 h after models establish-ment, while the control group received the same volume of physiological saline after models establishment. The neurologic impairment scores were detected at 24h after infarct, and the infarct volumes were also measured. Results The values of neurologic impairment scores and infarct volumes in group A, B and C were significantly lower than those in group E re-spectively (P<0. 05, P<0. 01), and the differences between group B and E were more significant (P<0. 01), and the values in group B were lower than those in group A, C and D respectively (P<0. 05, P<0. 01), and the differences be-tween group B and D were more significant ( P<0. 01 ) . Conclusion Edaravone has neuroprotective effect in MCAO rats, and the effect has the golden hour.
