目的 探讨移居高原早期血液中性粒细胞的线粒体形态学变化.方法 分别在海拔1400 m某地选取3名拟乘汽车进入海拔4200 m高原的健康男性青年,采集空腹静脉血5 ml(0 d),分离白细胞,戊二醛固定,采用透射电镜观察其中性粒细胞中的线粒体形态学变化.跟踪该3人到达4200 m高原,分别在到达高原后7、30 d时再观察上述指标.同时选取职业、年龄相匹配的3名从平原移居4200 m 高原1年的健康男性,测定其上述指标.结果 移居高原7 d时,线粒体核周间隙增宽,不均匀,线粒体数量较多,较多线粒体出现固缩、水肿等;移居高原30 d时,大量线粒体固缩,核周间隙扩张;移居高原1年时,线粒体数目较多,形态逐渐恢复正常,仅个别线粒体出现水肿.结论 移居高原早期线粒体的形态发生改变,但随着时间推移,线粒体的形态逐渐正常.%Objective To investigate the early morphologic changes of netrophil mitochondria in healthy young men after immigration into high altitude. Methods Three young men lived in 1400 m altitude who would arrive in a plateau of 4200 m altitude by automobile were enrolled in this study. Their fasting venous blood of 5 ml was collected on day 0, and then white blood cells were isolated and fixed by glutaraldehyde for the observation of morphologic changes of neutrophil mitochondria under transmission electron microscope. After these three men arrived at 4200 m altitude for 7 d and 30 d,the above morphologic indexes of their mitochondria was respectively observed. Meanwhile, the same morphologic indexes of other three young men who had migrated to 4200 m altitude for one year from the plain with matched occupation and age were observed as controls. Results On day 7 after migrating at high altitude,the perinuclear space of mitochondria became widened and uneven. There were many mitochondria in neutrophils and most of them were pyknotic and edematus. On day 30,a large number of pyknotic mitochondria were seen with expanded perinuclear space. One year after migration,most appearances of mitochondria had already recovered to normal, but less of them still showed edema. Conclusion Morphologic changes of neutrophil mitochondria can be found early after high - altitude migration. However, its appearance gradually becomes normal with the acclimatization to high altitude environment.
