单核细胞趋化蛋白-1诱导入主动脉平滑肌细胞表达基质金属蛋白酶-9及相关通路研究

         

摘要

To observe the effect of monocyte chemotactic protein-1 (MCP-l) on the expression of matrix metalloproteinase-9 (MMP-9) in human aortic smooth muscle cells (HASMCs) and the related signaling pathways. Methods The HASMCs were divided into two parts. In the first part, 0 (control group), 10, 50, and 100ng/ml MCP-l were respectively added to interfere with MMP-9 expression for 48h: then, Western blot analysis was used to detect the protein expression of MMP-9 and NF-KB p6S in the HASMCs. In the second part, l00ng/ml MCP-l was initially added, and then, 0 (control group), 10, SO, and 100u.mol/L of PDTC, as an NF-KB-specific inhibitor, was added to the cells. The cells were then cultured for 48h. Western blot analysis was used to detect MMP-9 expression in the HASMCs. Results The MMP-9 and NF-KB p6S expression in the HASMCs were dose-dependently stimulated by MCP-l. Compared with the control group, the addition of 10, 50, and lOOng/ml MCP-l upregulated MMP-9 expression by 70.0%, 118.3%, and 125.0% (P<0.0l), and NF-KB p65 protein expression by 32.0%, S2.0%, and 89.0%, respectively. The addition of 10,50, and 100μmol/L PDTC significantly downregulated MMP-9 expression by 22.1%, 28.9%, and 36.7%, respectively (P<0.01). Conclusion MCP-l induces MMP-9 expression in HASMCs. The NF- K B pathway regulates MCP-l expression to stimulate MMP-9 expression.%目的 观察单核细胞趋化蛋白-1(MCP-1)对人主动脉平滑肌细胞(HASMCs)基质金属蛋白酶-9(MMP-9)蛋白表达的影响及其相关机制.方法 将HASMCs细胞分为两部分,一部分分别加入0(对照组)、10、50、100ng/ml MCP-1干预48h,采用Western blotting法测定HASMCs细胞MMP-9、NF-kB p65的蛋白表达;另一部分先加入100ng/mlMCP-1,再分别加入0(对照组)、10、50、100μ mol/L的NF-kB特异性抑制剂吡咯醛二硫氨基甲酸(PDTC),培养48h,采用Western blotting法检测HASMCs细胞MMP-9蛋白的表达情况.结果 MCP-1以剂量依赖方式刺激HASMCs细胞内MMP-9蛋白和NF-kB p65蛋白的表达,与对照组比较,加入10、50、100ng/ml MCP-1可使MMP-9蛋白的表达分别增加70.0%、118.3%和125.0%,使NF-kB p65蛋白的表达分别增加32.0%、52.0%和89.0%(P<0.01).加入PDTC后,与对照组比较,10、50、100μmol/L的PDTC分别使MMP-9蛋白的表达降低了22.1%、28.9%和36.7%,均有显著性差异(P<0.01).结论 MCP-1可诱导HASMCs表达MMP-9.NF-kB通路是MCP-1刺激MMP-9表达的调控通路之一.
