PM2.5对儿童骨髓基质细胞增殖及细胞因子G-CSF、GM-CSF分泌的影响

     

摘要

Objective:To observe the effects of ambient particulate matter (PM2.5) on the proliferation and secretion levels of cytokines of children’s bone marrow stromal cells (BMSCs) and to explore the effects of PM2.5 on children’s bone marrow hematopoietic microenvironment.Method:The collected BMSCs were exposed to different concentration of PM2.5 solutions.The proliferation of BMSCs and the secretion levels of G-CSF, GM-CSF and mRNA were detected.Result:The BMSCs proliferation increased with the increase of concentration of PM2.5 solution,after 50μg/mL the proliferation of BMSCs level gradually decline.The proliferation of BMSCs level with concentration 20μg/mL of experimental group was significantly higher than control,while 100μg/mL of experimental group was significantly lower than control group,the differences were statistically significant (P<0.05).The low concentration of PM2.5 with 10 and 20μg/mL promoted the secretion levels of G-CSF, GM-CSF and mRNA,while gradually inhibited in the PM2.5 concentration of 50,100 and 200μg/mL.The concentration reaches more than 100μg/mL,inhibiting effects reached significant level.Conclusion:The high concentration of PM2.5 has a certain toxic effect on children’s BMSCs.%目的:研究大气细颗粒物(PM2.5)对儿童骨髓基质细胞(BMSCs)增殖及细胞因子分泌的影响,探讨PM2.5对儿童骨髓造血微环境的影响。方法:采用不同浓度的PM2.5溶液染毒提取后培养的BMSCs,检测BMSCs增殖和粒细胞集落刺激因子(G-CSF)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)分泌量及mRNA表达水平。结果:随着PM2.5溶液浓度增加,BMSCs增殖逐渐增强,溶液浓度达到50μg/mL后BMSCs增殖水平逐渐下降,20μg/mL时试验组BMSCs的OD值各时间段均高于对照组,而100μg/mL时均低于对照组,两组比较差异均有统计学意义(P<0.05)。低浓度(10、20μg/mL) PM2.5显著促进BMSCs分泌G-CSF、GM-CSF及mRNA表达,高浓度(50、100、200μg/mL)PM2.5抑制BMSCs分泌G-CSF、GM-CSF及mRNA表达,且浓度达到100μg/mL以上时,抑制效果达到显著水平。结论:高浓度PM2.5对儿童BMSCs具有一定的毒性作用。
