量子点荧光探针检测肺腺癌组织中EGFR的表达

         

摘要

Objective To investigate the expression of EGFR antigen in lung adenocarcinoma tissue samples which was detected by quantum dots (QDs) fluorescence probe.Compared with immunohistochemical method (IHC),we explored the application value of QDs fluorescent probe in the detection of specific markers in lung adenocarcinoma.Methods QDs fluorescent probe (prepared by aqueous phase method) and IHC method were both used to detect the expression of EGFR in lung adenocarcinoma tissues.Statistical analysis was performed using SPSS.Results The detection results of the two methods were of high consistency with the positive rate of EFGR in lung adenocarcinoma was as high as 96.6% (0.25 < Kappa < 0.75,P < 0.001).The strong positive rate (3 +) was significantly independent of TNM stage and grade.Conclusion QDs fluorescent probes can accurately label antigen expression intensity of the tissue samples in different stages and grades.The fluorescence intensity of the method is stable,and it can be applied to detect the expression of EGFR in lung adenocarcinoma.%目的 利用量子点(QDs)荧光探针检测肺腺癌组织样本中的EGFR抗原的表达,与免疫组化法(IHC)比较,探讨QDs荧光探针在肺腺癌组织中特异性标志物检测的应用价值.方法 水相法制备QDs荧光探针,并与IHC法分别检测肺腺癌组织中EGFR表达情况,利用SPSS进行统计学分析.结果 两种方法的检测结果具有较高一致性(0.4< Kappa<0.75,P<0.001),EFGR在肺腺癌中的阳性率高达96.6%,其中强阳性(3+)率与TNM分期、镜下组织形态分级显著无关(P>0.05).结论 QDs荧光探针能够精确标示不同分期、分级的组织样本中抗原表达强度,荧光强度稳定,可应用于检测肺腺癌中EGFR的表达.
