蝎毒素对小鼠多种免疫细胞活化的影响

摘要

Aim: To investigate the effects of buthus martensii karsch immune active (BMK IA) on activation of multiple immunocytes in mice. Methods: A total of 48 C57BL/6 mice were allocated into experimental group and control group. The mice in experimental group were injected intravenously with a single dose of BMK 1A(0. 004 mg/kg) ,and those in control group were given normal saline. The expression of cell-specific antigens and CD69 in peripheral monocytes, NK cells, B cells, CD4 + T and CD8 + T cells were assessed using immunofluorescent antibody labeling and flow-cytometry. The activation ratio was assessed. Results:BMK IA significantly stimulated and activated multiple immunocytes except CD4+T cell(P< 0.05). CD69+ percentages peaked within 8 ~ 12 h [ monocytes (44. 8 ± 19. 2)% ,8 h;NK cells(56. 2 ± 17. 2)% , 12 h;B cells (53.3 ±21.4)% ,8 h;CD4+T cells ( 14.4 ±5. 1)% ,12 h;CD8+T cells (63. 2 ±4. 5)% , 12 h] after BMK IA injection. Conclusion:BMK IA could enhance the immune function of mice,and is an effective biological response modifier.%目的:探讨东亚钳蝎毒素(BMK IA)对小鼠体内多种免疫细胞活化的影响.方法:C57BL/6小鼠48只,随机数字表法分为空白对照组和实验组,每组分为4批,每批6只,各检测一种免疫细胞.实验组小鼠一次性静脉注射0.004 mg/kg的BMK IA,对照组给予等体积的生理盐水.流式细胞术分别检测2组小鼠给药0、2、8、12和24h后外周血单核、NK、B、CD4+T和CD8+T细胞标志性抗原以及早期活化标志CD69的表达.结果:实验组小鼠外周血多种免疫细胞产生活化(P＜0.05),细胞活化率在8～12 h达到高峰[单核细胞(44.8±19.2)%,8 h;NK细胞(56.2±17.2)%,12 h;B细胞(53.3±21.4)%,8 h;CD4+T细胞(14.4±5.1)%,12 h;CD8+T细胞(63.2±4.5)%,12 h],CD4+T细胞活化不明显.结论:BMK IA可提高小鼠机体的免疫功能,是一种有效的生物应答调节剂.
