吡格列酮对哮喘小鼠肺组织 GABAARα、MUC5AC表达的影响

     

摘要

Aim:To observe the effects of pioglitazone on the expressions of GABAARαand MUC5AC in lung tissue of asthmatic mice, and to explore new ideas for prevention and treatment of asthma .Methods:Thirty BALB/C mice were randomly allocated into 3 groups(n=10):normal control group,the asthma group,and the pioglitazone intervention group . The latter 2 groups were established asthma model by sensitized and challenged with ovalbumin .The pioglitazone interven-tion group was given aerosol inhalation of pioglitazone 30 min before each allergen challenge .The pathological changes of the lung tissue were observed by HE staining .The expressions of GABAARα, MUC5AC mRNA and protein in lung tissue were analyzed by RT-RCR and immunohistochemistry .Results:The mice in the asthma group showed mucosal hyperemia and edema , mucosa thickening and severe airway inflammation .These changes alleviated in the pioglitazone intervention group.The expressions of GABAARα,MUC5AC protein and mRNA in the asthma group were higher than those of normal control group(P<0.05),and compared with those of the asthma group , the above indexes in the pioglitazone intervention group decreased but still higher than those in normal control group (P<0.05).Conclusion:Pioglitazone may play a role in the treatment of excessive mucus production through down-regulating the expression of GABAARα.%目的:观察吡格列酮对哮喘小鼠肺组织GABAARα、MUC5AC表达的影响。方法:30只成年BALB/C小鼠随机分为3组,每组10只。哮喘组和吡格列酮干预组以卵白蛋白( OVA)致敏、激发制作哮喘小鼠模型,吡格列酮干预组于每次激发前30 min雾化吸入吡格列酮5×10-5 mol/L,对照组以生理盐水代替OVA处理。末次激发后24 h处死小鼠,取肺组织行HE染色观察气道病理学表现,采用免疫组化SP法、RT-RCR法检测肺组织GABAARα、MUC5AC蛋白和mRNA的表达。结果:哮喘组小鼠气道黏膜下充血水肿,黏膜层增厚,支气管壁及血管壁周围有较多炎性细胞浸润;吡格列酮干预组上述表现较哮喘组减轻。哮喘组小鼠肺组织GABAARα、MUC5AC蛋白和mRNA的表达水平均较对照组高,吡格列酮干预组较哮喘组降低,但仍高于对照组(P均<0.05)。结论:吡格列酮可能通过下调GABAARα的表达在哮喘气道黏液过度分泌中起治疗作用。
