百会穴长留针疗法对智力偏低大鼠学习记忆能力影响的实验研究

         

摘要

[目的]观察百会穴长留针疗法对智力偏低大鼠学习记忆能力的影响,为临床上治疗智力相对偏低人群筛选出一种有效、方便的治疗方法,并为百会穴长留针疗法治疗智力相对偏低人群提供有效的实验依据.[方法]将170只SD大鼠通过Morris水迷宫进行筛选、分组,设立低智对照组、高智对照组、百会穴长留针组和西药对照组四个组.每10天用Morris水迷宫测试一次,观察3次.实验结束后用比色法测海马组织中ChAT、AchE活力.[结果]①在Morris水迷宫实验中,观察发现治疗30天后,与低智对照组比较,百会穴长留针组和西药对照组能显著缩短定位航行试验的潜伏期(P<0.01);②在空间探索试验中,与低智对照组比较,百会穴长留针组和西药对照组能提高跨越原平台位置的次数(P<0.05);③与低智对照组相比,百会长留针组、西药对照组能升高脑组织ChAT活力,其差异具有统计学意义(P<0.01);④与低智对照组相比,百会穴长留针与西药对照组均能抑制脑组织中AchE活力(P<0.01,P<0.05).[结论]百会穴长留针可以改善大鼠学习记忆能力,其机制可能是通过提高ChAT的活力,增加Ach含量;降低AchE的活性,减少Ach消耗,进而改善大鼠学习记忆能力.%[Objective] Observe the effects of long-time-retaining needle therapy at Baihui on the rats with relative low intelligence of learning and memory. [Methods] 170 Sprague-Dawley (SD) rats were screened through the Morris water maze, set up four groups: low intellectual control group, high intellectual control group, long-time-retaining needle at Baihui and comparison group. Morris water maze test was observed 3 times with every 10 days. After the experiment, measured by colorimetry acetylcholine transferase (ChAT), and acetylcholinesterase (AchE) activity. [Results] ①In the Morris water maze test, Compared the low intellectual control group after treatment for 30 days, the long-time-retaining needle at Baihui group and comparison group, can significandy shorten the latency of place navigation test(P<0.01);②Experiments in space exploration, compared the low intellectual control group, the long-time-retaining needle at Baihui group and comparison group can increase the number of times across the former platform location (P<0.05); ③Compared the low intellectual control group , the long-time-retaining needle at Baihui group and comparison group, ChAT activity in brain tissue could be increased, the difference was statistically significant(P<0.01);④Compared the low intellectual control group , the long-time-retaining ne-edle at Baihui group and comparison group could decrease the AchE activity in brain tissue (P<0.01, I°<0.05).[Conclusion] the long-time-retaining nee-die at Baihui can improve learning and memory in rats, which may be by increasing the vitality of ChAT and Ach content; reducing the activity of AchE and Ach consumption, improving learning and memory in rats.
