TCF7L2基因 rs290487C/T 多态性与新疆维吾尔族、汉族2型糖尿病的相关性

         

摘要

Objective To explore the correlation between rs290487C/T polymorphism of TCF7L2 gene and type 2 diabetes in Xinjiang Han and Uyghur populations.Methods Polymerase chain reaction and DNA sequencing were used to determine the genotype of rs290487C/T in 242 Chinese Han (116 in case group and 126 in control group)and 234 Chinese Uyghur (96 in case group and 138 in control group).Weight, height,waist circumference,blood pressure,fasting glucose,2 hour postprandial glucose,HbA1c,total cholesterol,high density lipoprotein cholesterol,low density lipoprotein cholesterol and triglycerides were measured.Results In Uyghur population,diabetes group showed significantly higher levels of BMI (P =0.000),TG (P =0.000),TC (P =0.013)and lower levels of HDL-C (P =0.001)than control group. The frequency of T allele was higher in Han people compared with Uyghur people (0.583 VS 0.348,P =0.000).For both Chinese Han and Uyghur,the frequencies of genotypes and particular allele in patients with T2DM showed no significant difference from those of the control group.In Uyghur,BMI of individu-als with CT or TT genotypes was significantly higher compared with that of the CC genotype (P <0.05). Conclusion The rs290487C/T polymorphism showed a racial and ethnic difference.No correlations be-tween rs290487 and type 2 diabetes in Xinjiang Han people and Uyghur people were found,but the rs290487 C allele may affect the BMI.%目的:探讨 TCF7L2基因 rs290487C/T 多态性与新疆维吾尔族、汉族2型糖尿病的关系。方法使用微测序法对新疆汉族242例(患者116例,对照126例)和维吾尔族234例(患者96例,对照138例)TCF7L2基因 rs290487C/T 多态性进行分析。同时进行生理生化指标的测定。结果维吾尔族糖尿病组 BMI (P =0.000)、TG (P =0.000)、TC (P =0.013)高于对照组,HDL-C 低于对照组(P =0.001)。汉族和维吾尔族正常人群TCF7L2基因 rs290487位点基因型频率和等位基因频率均存在统计学差异(P =0.000)。汉族和维吾尔族糖尿病组和对照组 TCF7L2基因 rs290487位点基因型频率和等位基因频率分布均无统计学差异(P >0.05)。维吾尔族糖尿病组和对照组中,携带 CC 基因型个体 BMI 显著低于携带 CT+TT 基因型个体(P =0.026,P =0.002)。结论TCF7L2基因 rs290487位点多态性存在种族和民族差异。TCF7L2基因 rs290487位点多态性与新疆汉族和维吾尔族2型糖尿病无相关性,但与 BMI 相关。
