辛伐他汀促进骨质疏松症大鼠种植体骨结合的实验研究

     

摘要

目的 探讨辛伐他汀对骨质疏松症大鼠种植体骨结合的影响.方法 将63只Wistar大鼠随机分为正常对照组、单纯去势组、去势加辛伐他汀组3组,通过手术去除双侧卵巢复制大鼠骨质疏松症模型,术后3个月,于每只大鼠左侧股骨处植入1枚种植体,1 d后各组分别灌喂生理盐水及辛伐他汀(5 mg·kg-1·d-1),术后4、8、12 w每组处死5只大鼠,取材制作非脱钙骨磨片,行骨组织形态学观察及骨计量学检测.结果 种植12 w时,单纯去势组种植体与周围骨组织有一定厚度新骨形成,与种植体接触面积增大.去势加辛伐他汀组和正常对照组种植体周围新生骨组织连接成片且厚度增加,与种植体表面紧密接触.种植术后4、8、12 w时,单纯去势组种植体骨结合率(IBCR)、新生骨量(NBV)均明显低于去势加辛伐他汀组和正常对照组,差异有统计学意义(P<0.01);去势加辛伐他汀组IBCR和NBV与正常对照组接近,差异无统计学意义(P >0.05).结论 辛伐他汀能明显增加骨质疏松症大鼠种植体骨结合率及种植体周围新骨形成量.%Objective To explore the effect of simvastation on osseointegration of implant in ovariectomized rats with osteoporosis.Methods Sixty-three female Wistar rats were randomly divided into three groups: sham operation group,ovariectomized group(OVX)and OVX with simvastatin treatment group.Postmenopausal osteoporosis rat model was established by removing ovaries,post-operation for 3 months,the pure titanium screws were implanted on each rat.One day after surgery,OVX with simvastatin treatment group(with simvastatin 5 mg·kg-1·d-1 for 12 weeks).The rats of other groups were fed with saline.Five rats from each group were euthanatized on the end of the 4th,8th,and 12th w respectively and the de-calcified sections were prepared and examined histologically and histomorphometrically.Results At 4th,8th and 12th week after implantation surgery,implant-bone contact rate(IBCR)and new bone volume(NBV)in ovariectomized group were all significantly decreased compared to sham operation group and OVX with simvastatin treatment group(P<0.01),while the bone indices(IBCR,NBV)in OVX with simvastatin treatment group were similar with those of sham operation group(P>0.05).Conclu-sion Simvastatin can significantly increase the rate of implant-bone contact rate and new bone volume in ovariectomized rats.
