和田羊KAP7、KAP8基因遗传多样性及其与羊毛长度的关联性分析

摘要

以采集的600只和田羊个体的血样为材料,利用 PCR-SSCP、DNA测序等技术研究分析和田羊KAP7、KAP8基因的多态性,并与羊毛长度进行关联.结果表明,和田羊KAP8基因没有发现多态性；KAP7基因具有多态性,存在3种基因型：AA、AB和BB,且处于 Hardy-Weinberg平衡状态(P＞0．05),多态信息含量为0．2376,属于低度多态.测序结果显示,与原基因碱基序列相比,KAP7基因碱基序列在110(T→G)和268(G→A)处发生突变.关联分析表明,和田羊不同基因型之间羊毛长度差异显著.%The blood samples of 600 Hetain sheep were applied as test materiats and PCR-SSCP,DNA se-quencing technology were used to research and analyze the genetic polymorphism of KAP7,KAP8 and carry out correlation with wool length.The result showed that there were no polymorphism in KAP 8 of Hetain sheep;However,there were polymorphism in KAP 7 gene of Hetain sheep,there were 3 genotypes:AA,AB and BB,and in the Hardy-Weinberg-equilibrium state (P>0.05),polymorphism information con-tent (PIC)was 0.2376,belonging to moderate polymorphism.Sequencing result showed that compared with the original genetic base sequence,KAP7 genetic base sequence was in 110G(T→G)and 268(G→A)point mutation.Correlation analysis showed that there were significant differences between different genotypes of Hetain sheep.
