交联纤维素酶聚集体制备及其催化特性研究

             

摘要

Cellulase was precipitated by ammonium sulfate and then cross-linked with glutaraldehyde, as a result, cross-linking cellulase aggregation was obtained. Finally, the optimum temperature, optimum pH, thermal stability, pH stability and batch cycle numbers of immobilized enzyme were studied. The highest enzyme activity obtained was 559. 2 U/g wet carriers. The optimum pH of the immobilized enzyme was 9.0, and the optimum temperature was 60°C, almost no activity lost after 10 cycles. Furthermore, cross-linking cellulase aggregation had better thermal and pH stability than free enzyme. Cross-linking cellulase aggregation had good operating stability, which had the potential for industrial biocatalyst use.%用90%饱和度的硫酸铵盐析沉淀纤维素酶蛋白,将蛋白沉淀用3%戊二醛交联固定制得交联酶聚集体;研究交联酶聚集体的最适反应温度、最适反应pH、酸碱稳定性、热稳定性、批次稳定性.交联酶纤维素酶聚集体的最高比活性为559.2 U/g湿载体,最适反应温度为60C,最适pH为9.0.交联酶聚集体的热稳定性和pH稳定性均优于游离酶,重复使用10批活性基本稳定.交联纤维素酶聚集体稳定性较好,具有一定的工业应用前景.
