目的 探讨压力性尿失禁( SUI)患者阴道前壁中Fibulin-1与SUI发生的关系.方法 采用实时荧光定量-PCR、免疫蛋白印迹方法测定25例压力性尿失禁患者(SUI组)阴道前壁组织中Fibulin-1的蛋白及mRNA表达水平,择同期27例非卵巢功能性肿瘤和宫颈病变患者作为对照(对照组).结果 RT-PCR结果显示Fibulin-1mRNA表达水平△Ct值在SUI组织(0.456 5±0.220 6)明显低于对照组组织(0.246 8±0.5017) (F =0.241,P<0.001);Fibulin-1 mRNA表达水平△Ct值在SUI绝经前组组织(0.373 2±0.158 2)明显低于绝经前对照组组织(1.157 5±0.1542)(F=0.212,P<0.001);Fibulin-1 mRNA表达水平△Ct值在SUI绝经后组组织(0.557 3±0.2219)与绝经后对照组组织(0.5326±0.2221)都很微弱,(F=1.099,P>0.05).结论 SUI患者的盆底支持结构的退行性病变与组织中的Fibulin-I低下有关.%Objective To investigate the expression of Fibulin-1 in anterior vaginal tissue and its relation to female stress urinary incontinence (SUI). Methods Tissue samples of anterior vaginal wall were taken from 25 women with SUI and uterine tissue samples were taken from 27 patients of non-ovarian tumors and cervical lesions as control group. The expression of Fibulin-1 mRNA and protein were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting respectively. Results The expression of Fibulin-1 mRNA 1( ΔCt)in premenopausal SUI patients was 0.373 2 ±0.158 2, which was lower than that of control premenopausal patients(0.157 5 ±0. 154 2, P < 0.001); the ACt value in SUI group was 0.456 5 ±0.220 6, which also lower than that of control group(0. 246 8 ± 0. 501 7, P < 0. 001). There was no significant difference in protein and mRNA expression between postmenopausal SUI patients and control group (P > 0. 05). Conclusion The degeneration of the pelvic floor support structure in SUI patients might be associated with the down-regulation of Fibulin-1.
