MYH9和ABI2基因SNP在2型糖尿病人群中与血糖、胰岛素抵抗和血压的相关性

         

摘要

Objective: To investigate the relationship of MYH9, ABI2 gene single nucleotide polymorphisms (SNPs) and glucose, insulin resistance, and hypertension in type 2 diabetes patients. Methods: Eight SNPs in MYH9 gene and 4 SNPs in ABO gene were genotyped in 773 type 2 diabetes patients in Tianjin area by matrix-assisted laser desorption/ionization time-of-ilight mass spectrometry(MALDI-TOF MS). Plink was used for quantitative association studies and genotype interactions (epistasis) analyses. Results: Significant associations were found among MYH9 and ABI2 SNPs and insulin resistance phenotypes, including HbAlc, 2-hr glucose, fasting and 2 hr plasma insulin(P<0.05). Moreover, association was found among ABI2/MYH9 SNPs and affection status of hypertension(rs62183937 and rsl 1675251 in ABI2, P=0.000, P=0.008, respectively). Conclusion: It is suggested that polymorphisms in these 2 genes are associated with glucose, insulin resistance, and hypertension related phenotypes in type 2 diabetes patients.%目的:研究MYH9和ABI2基因的单核苷酸多态性(SNP)在2型糖尿病人群中与血糖、胰岛素抵抗及高血压的关系.方法:应用基质辅助激光解吸电离飞行时间质谱技术,对773例天津地区的2型糖尿病患者的MYH9基因的8个SNP位点和ABI2基因的4个SNP位点进行基因分型,以PLINK软件进行数量性状的关联分析及基因型交互作用分析.结果:MYH9和ABI2基因SNP位点与血糖和胰岛素抵抗表型存在较广泛的关联,包括糖化血红蛋白、2h血糖、空腹和2h胰岛素及胰岛素抵抗指数(P<0.05).ABI2基因SNP位点rs62183937(P=0.000),rs11675251 (P=0.008)等均与血压相关.结论:MYH9和ABI2基因的多态性可能与2型糖尿病患者的血糖、胰岛素抵抗相关表型及2型糖尿病合并高血压的发病相关.
