目的:在人的非小细胞肺癌细胞A549中通过降低K-Ras表达研究其在自噬中的作用.方法:磷酸钙方法转染K-RasshRNA,Western blot检测自噬标志物的变化.结果:在A549细胞中,通过K-Ras-shRNA慢病毒转染显著地降低了K-Ras 的蛋白水平,同时自噬标志物LC3Ⅱ水平降低,自噬调控分子Beclin-1水平降低.结论:K-Ras knockdown引起细胞自噬水平的降低.%Objective:To explore the contribution of K-Ras to autophagy after K-Ras knockdown.Methods:Calcium phosphate method and Western blot were used to transfect K-Ras-shRNA and to detect the changes of autophagy marker,respectively.Results:In A549 cells,protein level of K-Ras was reduced significantly by K-Ras-shRNA and autophagy marker LC3Ⅱ decreased after K-Ras knockdown,so did autophagy regulating molecule Beclin-1.Conclusion:K-ras knockdown can result in the reduction of autophagy level.
