干扰素-γ对脊椎后纵韧带骨化成纤维细胞Runx2、Osterix表达的影响

             

摘要

目的 探讨干扰素-γ(interferon-γ,IFN-γ)对脊椎后纵韧带骨化成纤维细胞中Runx2、Osterix表达的影响.方法 采集脊椎后纵韧带骨化症患者后纵韧带组织,用贴壁法进行体外原代培养,传代后使用IFN-γ(50 U/μL)处理脊椎后纵韧带骨化成纤维细胞.第7、14天,通过半定量RT-PCR测定Runx2和Osterix的mRNA表达,通过SDS-PAGE测定Runx2和Osterix的蛋白质表达.结果 IFN-γ抑制成纤维细胞中Runx2和Osterix的mRNA和蛋白质的表达.结论 Runx2和Osterix在椎后纵韧带骨化中具有重要作用,但其具体作用机制尚需进一步研究.%Objective To investigate the effects of interferon-γ (IFN-γ) on the expression of Runx2 and Osterix in ligament fibroblasts derived from patients with ossification of posterior longitudinal ligament (OPLL). Methods Tissue samples of posterior longitudinal ligaments were collected from confirmed OPLL patients, and then the ligament fibroblasts were cultured in vitro using adherent primary culture and passaged. After the ligament fibroblasts were treated with 50 U/μX IFN-γ for 7 and 14 d, expressions of Runx2 and Osterix mRNA and protein levels were determined by semi-quantitative RT-PCR and Western blotting, respectively. Results IFN-γ treatment inhibited expression of Runx2 and Osterix at mRNA and protein levels in the ligament fibroblasts. Conclusion RurvZ and Osterix play important roles in the process of OPLL, but the underlying mechanism needs further investigation.
