高温对人前列腺癌细胞株PC-3m和LNCaP生长影响的体外研究

         

摘要

Objective To study the effect of hyperthemia on growth of human prostatic cancer cell lines PC- 3m and LNCaP in vitro. Methods PC- 3m and LNCaP were exposed to high temperature at 43 ℃ or37℃ for different duration( 1 h or 2 h) , and cell survival inhibition rate was evaluated by colony formation assay and measurement of cell growth rate. Results When PC- 3m and LNCaP were expesed to 43 ℃ for 1 or 2 hours, their mean inhibition rates of colony formation were (30.8 ± 2.4) % and (56.6±14.7) %, and (41.3±12.6) % and (94.2 ± 6.3) %, respectively. There was high significant difference between both cell lines( P<0. 005 and P<0. 001 respectively). These were similar to the results obtained in the cell growth curves. Conclusion Results indicate that induced hyperthermia has a cytotoxic effect on human prostatic cancer cells, moreover, temperature and exposure time have an important impact on the cell inhibition rate. Hyperthermia as a potential u.seful adjunctive therapy for local control of prostatic cancer merits further investigation.%目的探讨高温对人前列腺癌细胞株PC-3m和LNCaP生长的影响。方法用成集落实验和细胞生长曲线分析癌细胞在43℃和37℃作用1 h或2 h后的生长抑制率。结果PC-3m在43℃作用1 h和2 h的平均集落形成抑制率分别为(30.8±2.4)%和(56.6±14.7)%;LNCaP分别为(41.3±12.6)%和(94.2±6.3)%,差异比较均具有极显著性意义(分别为P<0.005和P<0.001)。与细胞生长曲线测定结果一致。结论高温对人前列腺癌细胞株具有细胞毒性作用,温度和作用时间对癌细胞生长的抑制也有重要影响;热疗作为治疗前列腺癌的一种辅助疗法,值得进一步研究。
