酶联免疫检测技术在水环境雌激素快速筛选中的应用研究

         

摘要

This study aimed to determine concentrations of the typical estrogen 17β - estradiol in water samples collected from the Yangtze estuary and wastewater treatment plants using enzyme - linked immunosorbent assay ( ELISA) ,and illuminated the feasibility and suitability of this method verified by chemical measured concentrations.The results showed that the lowest detection limit of ELISA was 0.5 ng · L-1 and the coefficient of variance for tests was below 30% which indicated high sensitivity and repeatability.The E2 concentrations detected by ELISA were at the range of below 0.5 ng · L-1 to 0.7 ng · L-1 for samples in the Yangtze estuary and 2.5 ng · L-1 to 11.5 ng · L-1 for samples in wastewater treatment plants,which had a good correlation (R2 =0.9147) with the results obtained from LC - MS/MS.By comparing the two detection methods,the water amounts of sample preparation in ELISA were lower than those in LC - MS/MS,and ELISA method shortened detection time for every sample attributed to simultaneous determination of 43 samples using the 96-well microplates,which indicated that ELISA was a rapid,convenient and economical method for estrogens analysis.%采用酶联免疫(ELISA)检测技术,对长江口水样和污水处理厂水样中的典型雌激素雌二醇(E2)进行分析,并与化学检测浓度进行验证比较.结果表明:ELISA的检测限可低至0.5 ng·L-1,试验的变异系数小于30%,具有较高的灵敏性和良好的重复性;ELISA检测长江口采集的水样中E2浓度范围在0.5 ng· L-1以下到0.7 ng·L-1,污水处理厂水样中E2浓度范围在2.5~11.5 ng· L-1,与液相色谱串联质谱( LC - MS/MS)的分析结果相比,相关系数R2为0.9147,表明两种方法具有良好的相关性;ELISA检测所需的水样量比LC - MS/MS更少,使用96微孔板可同时对43个样品进行检测,从而缩短了单个样品的分析时间,具有快速简便和经济适用性.
