ACE I/D和 CYP11 B2-344 C/T基因多态性与齐齐哈尔汉族人群原发性高血压的关系

         

摘要

Objective To explore the polymorphism of ACE I/D and CYP11B2-344 C/T and the genetic interaction between ACE and CYP11B2 genes in Qiqhar essential hypetension (EH) Han population.Methods Four hundreds and eighty-six patients with EH and four hundreds and fifty-seven healthy controls were enrolled . Genotyping was performed using PCR-RFLP technique .Results ( 1 ) genotype distributions of ACE I/D ( II 0.344 vs 0.326, ID 0.372 vs 0.497, DD 0.284 vs 0.177,P<0.001) between EH group and NT group;there was significant differences in D allele frequency of ACE I /D (0.470 vs 0.404, P<0.05) between two group. (2) genotype distributions of CYP11B2-344 C/T ( CC 0.136 vs 0.166, CT 0.469 vs 0.438, TT 0.395 vs 0.385, P>0.05) between EH group and NT group;there was not significant differences in T allele frequency of CYP11B2-344 C/T (0.630 vs 0.615, P>0.05) between two group.(3) The genotype distributions of DD +CT of EH group was significantly higher than that of NT group (P<0.01), the genotypes distribution of ID +TT of EH group was significantly lower than that of NT group (P<0.05).Conclusions These observations suggest that ACE I/D polymorphism is associated with essential hypertension in Qiqihar Han population , CYP11B2-344 C/T polymorphism is not associated with essential hypertension in Qiqihar Han population , and there might be mutual effect between the two genes .%目的:观察齐齐哈尔地区原发性高血压( EH)患者ACE I/D和CYP11B2-344 C/T基因多态性及其在EH发病中的相互作用。方法 EH患者486例,血压正常者457例。取外周血提取基因组DNA,用PCR-RLFP对ACE I/D和CYP11B2-344 C/T进行基因分型。结果(1)高血压组和血压正常组ACE I/D各种基因型分别为II 0.344/0.326、ID 0.372/0.497、DD 0.284/0.177(P<0.001),D等位基因频率为0.470/0.404(P <0.05);(2)高血压组和血压正常组 CYP11B2-344 C/T 各种基因型分别为 CC 0.136/0.166、CT 0.469/0.438、TT 0.395/0.385(P>0.05),T等位基因频率为0.630/0.615(P>0.05);(3)高血压组DD+CT联合基因型频率高于血压正常组(P<0.01);而高血压组ID+TT联合基因型频率低于血压正常组( P <0.05)。结论 ACE I/D 多态性与齐齐哈尔地区人群的原发性高血压相关, CYP11B2-344 C/T基因多态性与齐齐哈尔地区人群的原发性高血压不相关;但两基因可能存在相互作用。
