Objective To explore the experimental research of CPPs -BDI-1-THP-Loaded albumin nanoparticle triad killing EJ human bladder cancer cells , in other to build the new drug delivery system of targeting for follow -up experiments and treatment of bladder cancer .Methods SPDP was use to decoupling league to build nanometer microspheres triplet CPPs -BDI-THP-NS with the preparation of TAT -EGFP, BDI-1-NS and THP-NS.Take CPPs -BDI-1-THP-NS triplets as experimental group and THP as control group to detect the kill effect on EJ human bladder cancer cells by MTT method and detecting bladder cancer cells apoptosis by Annexin V-FITC/PI method.Results The inhibition effect of CPPs -BDI-1-THP-NS triplet killing EJ human bladder cancer cells was obviously stronger than the control group .Conclusions CPPs-BDI-1-THP-NS triplets has an obvious killing effect on human bladder cancer cells .%目的 探讨CPPs-BDI-1-载吡柔比星白蛋白纳米微球三联体杀伤人膀胱癌细胞研究,为后续实验构建靶向膀胱癌的新型药物传送系统及治疗奠定基础. 方法 采用异型双功能连接剂SPDP耦联穿膜肽增强型绿色荧光蛋白(TAT-EGFP)、抗人膀胱癌单克隆抗体(BDI-1)、载吡柔比星白蛋白纳米微球( THP-NS) ,构建载吡柔比星白蛋白纳米微球三联体( CPPs-BDI-1-THP-NS). 取CPPs-BDI-1-THP-NS三联体实验组和吡柔比星对照组,通过MTT法检测CPPs-BDI-1-THP-NS三联体对EJ人膀胱癌细胞的杀伤效果;Annexin V-FITC/PI法检测膀胱癌细胞的凋亡情况. 结果 CPPs-BDI-1-载吡柔比星白蛋白纳米微球三联体实验组杀伤人膀胱癌细胞作用明显强于对照组.结论 CPPs-BDI-1-载吡柔比星白蛋白纳米微球三联对人膀胱癌细胞有明显的杀伤作用.
