吴茱萸碱抑制涎腺腺样囊性癌细胞系ACC-M增殖的实验研究

         

摘要

Objective: To evaluate the effects of evodiamine on the proliferation of salivary adenoid cystic carcinoma ACC-M cells. Methods: ACC-M cells were treated with evodiamine at 0. 01, 0. 1, 1, 10 and 100 μg/ml for 24, 48 and 72 h respectively. MTT assay, phase contrast microscope, Wright-Giemsa staining and flow cytometry with Annexin-V-FITC/propidium iodide (PI) double staining were used to examine the growth inhibition and to observe the apoptosis of ACC-M cells. Results: Evodiamine inhibited the proliferation of ACC-M cells in a dose-and-time dependent manner, the IC50 values of evodiamine after 24, 48 and 72 h treatment were 6. 5 , 4.5 and 0.85 μg/ml respectively. The highest inhibition rate was 89%. Evodiamine induced apoptosis and necrosis of ACC-M cells. Conclusion; Evodiamine may inhibit the proliferation of ACC-M cells by induction of apoptosis and/or necrosis of the cells.%目的:研究吴茱萸碱对涎腺腺样囊性癌ACC-M细胞增殖的影响.方法:用终浓度为0.01、0.1、1,10,100 μg/ml的吴茱萸碱处理ACC-M 24、48、72 h后,应用MTT比色试验、倒置显微镜、Giemsa染色、Annexin-V-FITC和流式细胞仪检测和观察ACC-M的生长抑制率和凋亡情况.结果:吴茱萸碱(1 μg/ml以上)对ACC-M细胞具有时间-浓度依赖性生长抑制作用,最大生长抑制率可达89%.吴茱萸碱可诱导ACC-M细胞凋亡,凋亡率随处理时间延长和药物浓度增加而上升,各组间有显著差异.吴茱萸碱同时可引起ACC-M细胞坏死.结论:吴茱萸碱可以诱导细胞凋亡和/或坏死,从而抑制涎腺腺样囊性癌ACC-M 细胞增殖.
