用ELISA法检测单肺通气不同时间后大鼠血管内皮生长因子及可溶性E选择素的水平

         

摘要

Objective To study the changes of vascular endothelial growth factor (VEGF) and soluble E-selectin (sE-selectin) after different duration of one lung-ventilation (OLV) in rats. Methods Thirty SD rats were randomly divided into 5 groups (each n = 6): control group received double lung-ventilation. Group 1, 2, 3, and 4 received OLV for 1/2 h, 1 h, 2 h, and 4 h, respectively, with tracheal catheter inserted into right lung. The levels of TNF-a and IL-8 in lung tissues were determined, and the levels of sE-selectin in serum and VEGF in serum and lung tissues were determined by sandwich enzyme-linked immunosorbent assay (ELISA). Ultrastructural changes of lung tissues were observed by electric mirror. Results The levels of TNF-a and IL-8 in lung tissues were gradually increased along with the time of OLV. The levels of sE-selectin in serum in Group 2 , 3, and 4 were significantly increased than that in control group (P < 0.05). The level of VEGF in lung tissues in Group 4 was decreased as compared with that in control group (P < 0.05), while that in serum was increased (P < 0.05) The ultrastructural changes of lung tissues were lamellar body emptied , mitochondria damaged in Group 1, 2, 3 , and 4. Conclusion Long time OLV could induce acute lung injury, in which VEGF and sE-selectin play an important role.%目的:探讨不同时间单肺通气后大鼠血管内皮生长因子(VEGF)及可溶性E选择素(sE选择素)表达的变化.方法:将30只SD大鼠随机分成5组各6只,对照组行双肺通气,01组、02组、03组、04组分别为将气管切开后将导管插入右侧主支气管行右侧肺单肺通气1/2、1、2.4 h,听诊确定左侧肺萎陷.实验结束后检测肺组织中TNF-α和IL-8含量;用ELISA法检测血浆中sE-选择素、肺组织和血浆中VEGF的表达;电镜观察肺组织超微结构变化.结果:肺组织中TNF-α和IL-8含量随着单肺通气时间延长而逐步升高;SE-选择素在大鼠单肺通气1、2、4 h血浆中的表达较正常组显著升高(P<0.05);VEGF在大鼠单肺通气4 h组肺组织中的表达较正常组显著降低(P<0.05),而在该组血浆中的表达较正常组显著升高(P<0.05);实验组电镜下细胞的超微结构改变,板层小体排空,线粒体损伤等改变.结论:长时间单肺通气可以导致急性肺损伤(ALI),sE-选择素及VEGF在大鼠ALI形成过程中起着一定的作用.
