ObjectiveTo study the effects of malaytea scurfpea fruit and fructus ligustri lucidi on the migration and c-KIT expression of mouse melanoma cell line (B16).MethodsB16 cells were treated with malaytea scurfpea fruit and fructus ligustri lucidi respectively, then the cell migration was detected by transwell assay, and c-KIT expression was detected by RT-PCR and western blot.ResultsMigrated cells in every vision of the three malaytea scurfpea fruit treated groups and the three fructus ligustri lucidi treated groups were more than the corresponding control group respectively, there was a signiifcant difference (P<0.05), and the number of the migrated cells was affected by the concentration of the medicine in a dose dependent manner. Compared with the control group, c-KIT expression of melanoma cells of malaytea scurfpea fruit group (1.0×10-5mmol/L) and fructus ligustri lucidi group (1.0 g/L) was increased respectively, the difference was statistically signiifcant (P<0.05).ConclusionMalaytea scurfpea fruit and fructus ligustri lucidi promoted mouse B16 melanoma cell migration by promoting its c-KIT expression, which may be the mechanism of their therapeutic effects on vitiligo.%目的:探讨中药补骨脂素和女贞子对小鼠B16黑素瘤细胞c-KIT表达的影响及意义。方法体外培养小鼠B16黑素瘤细胞,用反转录-聚合酶链反应(RT-PCR)和蛋白质印迹法(Western blot)测定补骨脂素和女贞子对小鼠B16黑素瘤细胞c-KIT表达的影响,用Transwell小室法检测补骨脂素和女贞子对小鼠B16黑素瘤细胞迁移功能的影响。结果补骨脂素与女贞子用药组迁移到膜下层的每视野细胞个数分别较对照组多(P<0.05),并呈剂量依赖关系;用药组c-KIT mRNA、c-KIT蛋白表达分别比对照组增加,差异均有统计学意义(P<0.05)。结论补骨脂素和女贞子可能是通过促进小鼠B16黑素瘤细胞c-KIT的表达而促进其迁移功能,从而对白癜风产生治疗作用。
