一种制备无菌松材线虫的方法

         

摘要

报道了一种通过用H2O2处理松材线虫卵进而获得无菌松材线虫的方法.实验结果表明,15%H2O2处理对松材线虫卵有一定影响,随着处理时间延长,松材线虫孵化率下降,处理30min以上使其孵化率降低至10%以下;15%H2O2处理时间对获得无菌松材线虫有较大影响,其中,处理60min后获得无菌线虫的机率达70%.在无菌条件下,用无菌松材线虫对赤松无菌组织培养幼苗进行接种能使其发生萎蔫,表明采用该方法制备无菌松材线虫并未使其失去致病性,是一种成功的方法.%This paper describes a method for obtaining aseptic pine wood nematode (Bursaphelenchus xylophilus) by egg disinfection. Results showed that 15 percent hydrogen peroxide had a profound effect on egg hatching of pine wood nematode. The hatching rate of the pine wood nematode decreased with the prolonged processing time, and it decreased to less than 10 percent by soaking the eggs in 15 percent hydrogen peroxide for more than 30 min. The obtained aseptic pine wood nematode was obviously influenced by egg processing time in 15 percent hydrogen peroxide, and the probabihty of obtaining aseptic pine wood nematode was up to 70 percent after processing for 60 min. lte microcuttings of Pinus densiflora were inoculated with aseptic pine wood nematode under aseptic conditions, which resulted in the wilt of microcuttings. It showed that the asepsis did not cause the pine wood nematode to lose its pathogenicity.
