观察高脂饲料诱发的C57BL/6J小鼠血脂、转氨酶和肝脏脂质的动态变化.C57BL/6J分成基础饲料组和高脂饲料组,在给予高脂饲料后5、7、9、11周采血检测血脂、转氨酶,并制成10%肝匀浆检测肝脏脂质.取肝脏HE染色、油红O染色作病理学检查.结果表明,给予高脂饲料5周小鼠血脂TC和ALT显著升高,7~9周稳定,11周稍有下降.肝脏显示明显病理学损伤,呈现轻至中度非酒精性脂肪肝.高脂饲料引起C57BL/6J小鼠血脂TC和ALT升高迅速、明显,适合非酒精性脂肪肝模型的制作.%To establish a non-alcoholic steatohepatitis animal model induced by high-fat diet, and observe the dynamic variation of relevant index at different time. C57BL/6J mouse were randomly divided into 2 groups. The control group was fed with standard diet and the model group fed with high-fat diet. The serum TG, TC, ALT, AST, and ALP were assayed at the 5th, 7th, 9th and 11th week. Some mice were killed and the liver were taken, and liver homogenate lipid were detected. HE-stained sections were prepared for histopathological examination. The levels of TC and ALT in serum rose rapidly after 5 week of high-fat diet, and stayed at a high level throughout the duration of 7-9 weeks, and 11 week decreased a little. Pathological changes were observed in the liver. Animal Models presented slight to moderately degree Non-alcoholic Steatohepatitis. Feeding with high-fat diet could induce hyperlipidemia in mice rapidly and reliably, thus this method fit the establishment of animal model in non-alcoholic steatohepatitis.
