目的:探讨转移相关基因2(MTA2)对喉癌细胞系Hep-2的增殖、迁移和侵袭的影响.方法:通过针对MTA2基因的小干扰RNA(siRNA-MTA2)对MTA2基因表达进行下调,采用RT-PCR和Transwell检测转染,作为实验组,对照组转染阴性对照(sicontrol),通过MTT、细胞划痕实验和Transwell法分别检测MTA2对Hep-2的增殖、迁移和侵袭的影响.结果:实验组MTA2mRNA和蛋白表达水平明显低于对照组(P<0.05),实验组培养7d和9d时细胞增殖能力明显低于对照组(P<0.05),实验组细胞迁移能力和细胞侵袭能力明显低于对照组(P<0.05).结论:MTA2与喉癌细胞系Hep-2细胞增殖、迁移和侵袭密切相关,有望成为治疗喉癌的靶点.%Objective:To investigate the effect of metastasis associated gene 2 (MTA2) on the proliferation,migration and invasion of laryngeal carcinoma cell line Hep-2.Methods:MTA2 gene expression was down regulated by small interfering RNA (siRNA-MTA2) of MTA2 gene,and the transfection was detected by RT-PCR and Transwell.As the experimental group,the control group was transfected with negative control (sicontrol),and the effect of MTA2 on the proliferation,migration and invasion of Hep-2 was detected by MTT,cell scratch test and Transwell assay.Results:the expression level of MTA2mRNA and protein in the experimental group was significantly lower than that in the control group (P<0.05).The cell proliferation ability of the experimental group was significantly lower than that of the control group (P<0.05) when cultured in 7d and 9D,The cell migration ability and invasion ability of the experimental group were significantly lower than those of the control group (P<0.05).Conclusion: MTA2 is closely related to the proliferation,migration and invasion of Hep-2 cell line,which is expected to be a target for the treatment of laryngeal cancer.
