大鼠单侧睾丸扭转/复位后对双侧睾丸生殖细胞凋亡及MDA水平的影响

         

摘要

目的 探讨大鼠单侧睾丸不同时间扭转、复位对双侧睾丸生殖细胞的影响以及可能机制.方法 选用健康雄性SD大鼠35只,随机分成假手术组、单纯扭转组及扭转复位组.单纯扭转组采用Turner方法 建立左侧睾丸扭转模型(左侧睾丸逆时针扭转720°),分别在扭转持续2、6、12h后即处死;扭转复位组在左侧睾丸扭转后2、6、12h再复位,复位后24h处死.留取双侧睾丸标本进行HE染色观察、生殖细胞凋亡检测及丙二醛(MDA)含量测定.结果 睾丸扭转2h,单纯扭转及扭转复位后与假手术组比较扭转侧生殖细胞凋亡增加(P<0.05),而对侧生殖细胞凋亡增加差异无统计学意义(P>0.05);双侧MDA含量均增加(P<0.05).随着扭转时间的延长,单纯扭转及扭转复位后细胞凋亡和MDA含量均明显增加(P<0.05).结论 随着扭转时间的延长,扭转复位后睾丸的损伤加重,其中缺血-再灌注的氧自由基损伤是造成睾丸损伤的可能机制之一.%Objective To investigate the germ cell apoptosis and MDA concentration following unilateral testicular torsion/detorsion and to explore the possible mechanism which maybe cause the changes of germ cell apoptosis. Methods Thirty - five healthy male rats were randomly divided into sham operation group, pure detorsion group and torsion/detorsion group. Left testes torsion model was established with Turner method in pure detorsion group. The torsion/detorsion group underwent untwisting in torsed testis and they were sacrificed 24h later. All testes underwent histological, germ cell apoptosis and MDA concentration examination. Results Compared with control group,the difference of germ cell apoptosis in reverse side was statistically significant( P <0. 05 ) after torsing 2h,but there were no statistically significant in the contralateral side(P >0. 05). The concentration of MDA increased significantly in both side. With the extension of time, the germ cell apoptosis and the MDA concentration increased significantly in each experimental groups (P <0. 05 ) . Cnclusion The different damage of germ cell apoptosis comes from the different situation of testicular torsion/detorsion. The damage is related to the ischemia reperfusion injury. The mechanism maybe related to free oxygen radical.
