人胃腺癌组织中RUNX3、EGFR、VEGF表达及其与淋巴转移和血管侵犯的关系

             

摘要

目的 观察RUNX3、EGFR、VEGF在人胃腺癌组织中的表达,探讨其在胃癌淋巴转移和血管侵犯中的作用.方法 采用免疫组织化学EliVision法检测122例人胃腺癌组织中RUNX3、EGFR、VEGF的蛋白表达水平,分析其与胃腺癌临床病理特征的关系,以及三者表达的相关性.Western blot检测胃癌与癌旁组织中RUNX3的表达.结果 122例人胃腺癌组织中RUNX3、EGFR、VEGF的阳性表达率分别为37.7%(46/122),50.82%(62/122),73.77%(90/122).不同年龄、性别、肿瘤部位、肿瘤大小及分化程度的患者胃腺癌组织中RUNX3、EGFR、VEGF表达差异均无统计学意义(P>0.05),而在肿瘤有无淋巴结转移和有无血管侵犯的患者胃腺癌组织中三者表达差异均有统计学意义(P<0.05).当伴有淋巴转移和血管内癌栓时,RUNX3的表达下降(P<0.05),EGFR和VEGF的表达升高(P<0.05或<0.01).相关分析显示,EGFR和VEGF的表达呈正相关(r=0.383,P=0.000),人胃腺癌组织中RUNX3与EGFR和VEGF呈负相关(r=-0.317或r=-0.344,P=0.000).胃癌组织中RUNX3相对表达量(0.1464±0.1063)低于癌旁组织(0.2080±0.1255)(t=-3.553,P=0.001).结论 RUNX3、EGFR、VEGF三者参与人胃腺癌的发生发展过程,主要体现为调节淋巴转移和血管侵犯.%Objective To investigate the expression level of RUNX3, EGFR, VEGF in gastric adenocarcinoma tissues and to explore their role in the progression of lymphatic metastasis and vascular invasion. Methods The protein level of RUNX3, EGFR,VEGF in 122 cases of gastric adenocarcinoma tissues were detected using immunochemical EliVision methods. Western blot was used to detect the protein level of RUNX3 in gastric cancer and their adjacent tissues. Results The positive expression of RUNX3, EGFR, VEGF in 122 cases of gastric adenocarcinoma tissues were 46/122(37.7%),62/122(50.82%),90/122(73.77%),respectively. RUNX3, EGFR and VEGF showed no significant difference in gastric adenocarcinoma patients with different age, gender, tumor location,tumor size and differentiation degree(P>0.05),whereas, they all had significant differences when the tumor tissues with lymph node metastasis and vascular invasion(P<0.05). With the presence of lymphatic metastasis and vascular tumor emboli existence, the expression of RUNX3 declined significantly (P<0.05), however, the expression of EGFR and VEGF ascended(P<0.05 or <0.01,respectively). Correlation analysis showed that the expression of EGFR and VEGF had significant positive correlation with each other (r=0.383,P=0.000), but RUNX3 had negative correlation with them(r=-0.317 or r=-0.344,P=0.000). The expression level of RUNX3 protein in gastric cancer tissues was significantly lower than that in adjacent tissues. Conclusion RUNX3,EGFR and VEGF participants in the development of gastric adenocarcinoma mainly lie in their potential ability to regulate tumor lymphatic metastasis and vascular invasion.
