三氧化二砷对胃癌NCI－N87细胞增殖的影响

摘要

Objective To investigate the effect of arsenic trioxide ( As2 O3 ) on the proliferation and apoptosis of gastric cancer cells NCI-N87.Methods NCI -N87 cells were treated with 50 umol/L of As2 O3 , after 24h, MTT assay was used to detect the in-hibition effect of As2 O3 on the proliferation of NCI-N87 cells, and cell apoptosis was detected by TUNEL.Results The proliferation rate of NCI-N87 cells after treated with 50 umol/L As2 O3 for 24h was 21.3%.As2 O3 could induce the apoptosis of NCI-N87 cells. In different regions the apoptosis rate of NCI-N87 cells after treated with As2 O3 for 24h was 37.8%、34.00%、35.40%、36.8%( P<0.01), the apoptosis rate of normal NCI-N87 cells for 24h was 6.20%, 5.00%, 7.60%, 7.00%( P <0.01)..Conclusion As2 O3 can induce the apoptosis of gastric cancer cells, which can inhibit the proliferation of gastric cancer cells NCI-N87.%目的：研究三氧化二砷（ arsenic trioxide，As2 O3）对胃癌细胞系NCI－N87的细胞增殖与凋亡的影响。方法用50umol／L的As2 O3处理NCI－N87细胞，培养24h后，用MTT法检测 As2 O3对NCI－N87细胞的增殖影响，和TUNEL法检测细胞经过As2 O3处理后细胞凋亡情况。结果50umol／L的As2 O3作用24h后NCI－N87细胞的细胞增殖率是21．3％。经过As2 O3处理的NCI－N87细胞相比没有进行加药处理的细胞出现了大量的细胞凋亡。 As2 O3作用24h后NCI－N87细胞的不同视野细胞凋亡率分别是37．8％、34．00％、35．40％、36．8％（ P ＜0．01），正常没有加药培养的NCI－N87细胞的细胞凋亡率是6．20％、5．00％、7．60％、7．00％（ P ＜0．01）。结论通过As2 O3诱导其细胞凋亡，使胃癌NCI－N87细胞的细胞增殖有明显的抑制作用。
