UCA1和miR-141在膀胱癌细胞系和膀胱癌组织中的表达及与临床预后的相关性

摘要

Objective To investigate the correlation between clinical prognosis and expressions of UCA1 and miR-141 in cells and tissues of bladder cancer.Methods The bladder urothelial carcinoma cell line and urothelial carcinoma tissues were selected as the subjects.Normal bladder urothelial cell line and paracancerous tissues were selected as the control groups.The expressions of UCA1 and miR-141 were detected with real-time quantitative PCR.The correlation between prognosis and the expressions of UCA1 and miR-141 was analyzed.Results Compared with the controls,bladder urothelial carcinoma cell line and tissues had significantly increased expressions of UCA1 and miR-141 (P＜0.05).The expression of UCA1 was significantly associated with pathological grade,clinical stage and tumor metastasis(P＜0.05).The expression of miR-141 was significantly correlated with clinical stage and tumor metastasis(P＜0.05).Cox regression analysis showed that clinical stage,tumor metastasis,UCA1 and miR-141 expressions were independent risk factors affecting the prognosis of patients.The expressions of UCA1 and miR-141 in bladder urothelial carcinoma tissues were positively correlated(r=0.613,P＜0.05).Conclusion UCA1 and miR-141 are both highly expressed in bladder cancer cells and tissues,and are closely related to the clinical stage,tumor metastasis and prognosis.%目的 探讨UCA1和miR-141在膀胱癌细胞和膀胱癌组织中的表达及与临床预后的相关性.方法 选取人膀胱尿路上皮癌细胞系及宜宾市第二人民医院收治患者的膀胱尿路上皮癌组织标本为研究对象,另外分别选取人膀胱尿道正常细胞系和癌旁正常组织为研究对照,采用实时荧光定量PCR检测UCA1和miR-141表达情况,分析UCA1、miR-141表达与临床参数及预后的关系.结果 与膀胱尿道正常细胞系和癌旁正常组织比较,膀胱尿路上皮癌细胞系及膀胱尿路上皮癌组织中UCA1和miR-141相对表达量均显著升高,差异具有统计学意义(P＜0.05).UCA1的表达与病理分级、临床分期及肿瘤转移显著相关(P＜0.05);miR-141的表达与临床分期及肿瘤转移显著相关(P＜0.05).Cox回归分析显示,临床分期、肿瘤转移、UCA1和miR-141表达是影响患者预后的独立危险因素.膀胱尿路上皮癌组织中UCA1和miR 141表达呈正相关(r=0.613,P＜0.05).结论 UCA1、miR-141在膀胱癌细胞和膀胱尿路上皮癌组织中均呈高表达,且与膀胱癌患者临床分期、肿瘤转移及预后效果密切相关.
