Objective :To compare the cytotoxic T lymphocyte( CTL ) activity stimulated in vitro by dendritic cells ( DC ) loaded with heat - shocked and freeze - thawed gastric cancer cell strain gastric cancer cell strain. Methods : DC isolated from peripheral blood mononuclear cell of 12 gastric cancer peoples were loaded with heat shock - induced apoptotic gastric cancer cell strain and freeze - thawed tumor cell lysates separately. Using lymphocytes stimulated in vitro by DC as effector cells and gastric cancer cell strain as target cells. The cell - killing effect of effector cells on target cells was determined by MTT method. Results : DC loaded with heat shock - induced apoptotic gastric cancer cell strain and freeze - thawed tumor cell lysates both showed the CTL activity on target cells, but the CTL activity of the former was significantly higher than the latter ( P < 0. 05 ). Conclusion : The method of loading with heat shock - induced apoptotic gastric cancer cell strain was more effective.%目的:比较热休克凋亡和冻融裂解人胃癌细胞株抗原负载树突状细胞(dendritic cell,DC)体外刺激细胞毒T细胞(cytotoxic T lymphocyte,CTL)的活性.方法:分别制备热休克诱导凋亡胃癌细胞株和肿瘤细胞冻融裂解物,同时分离12例胃癌患者外周血单核细胞体外诱导DC进行负载,并以两种不同方式负载的DC进行淋巴细胞增殖反应及CTL杀伤实验,比较其刺激淋巴细胞增殖及激活的淋巴细胞杀伤靶细胞能力的差异.结果:负载热休克胃癌细胞的DC与负载反复冻融肿瘤细胞裂解物的DC都显示对靶细胞CTL活性,但前者的CTL活性显著高于后者 (P<0.05).结论:用热休克肿瘤细胞负载DC是一个更为有效的负载方式,为临床应用提供了一种可选择的负载方式.
