5-脂氧合酶抑制剂对人肝癌细胞株HepG2增殖和凋亡的影响

         

摘要

目的:研究5-脂氧合酶(5-lipoxygenase,5-Lox)在人肝癌细胞株HepG2中的表达,以及5-Lox抑制剂对人肝癌细胞株HepG2增殖、凋亡的影响.方法:应用免疫细胞化学染色及RT-PCR分别检测5-Lox的蛋白和mRNA在人肝癌细胞株HepG2中的表达,采用MTT法及流式细胞术检测5-Lox抑制剂齐留通(Zileuton)对人肝癌细胞株HepG2增殖、凋亡的影响.结果:5-Lox蛋白表达于肝癌细胞浆,局灶细胞核膜亦见阳性染色,RT-PCR可检测到肝癌细胞5-Lox的mRNA的表达.人肝癌细胞株HepG2给予不同浓度的齐留通处理24小时后,细胞存活率呈剂量依赖性下降;流式细胞术检测可见细胞凋亡率呈剂量依赖性升高.结论:人肝癌细胞株HepG2存在5-Lox的表达,5-Lox抑制剂齐留通可抑制人肝癌细胞株HepG2的增殖,诱导细胞凋亡.%Objective : To examine the expression of 5 - lipoxygenase in human hepatoma cell line ( HepG2 ) , investigate the effect of its specific inhibitor Zileuton on cell proliferation and apoptosis. Methods :Immunochemical staining and RT - PCR were used to detect the expressions of 5 - lipoxygenase protein and mRNA. Cell proliferation and apoptosis were determined by MTT assay and flow cytometry. Results : 5 - lipoxygenase protein was detected in cytoplasm, with focal nuclear membrane staining. The mRNA of 5 - lipoxygenase was detected in HepG2 cells. After the treatment of Zileuton for 24 hours , Zileuton reduced cell proliferation and induced apoptosis in HepG2 cells in a concentration - dependent manner. Conclusion : 5 - lipoxygenase is expressed in human hepatoma cell line ( HepG2 ). Inhibition of 5 - lipoxygenase by Zileuton reduces cell proliferation and induces apoptosis in a concentration - dependent manner.
