miR -450a -5p 对人卵巢癌 SKOV3细胞生物学行为的影响

         

摘要

Objective:To investigate the effect of miR - 450a - 5p on proliferation,migration,invasion,cell cycle and apoptosis in SKOV3 cells and explore its effect on the biological behavior of serous ovarian cancer SKOV3 cells. Methods:SKOV3 cells were transfected with miR - 450a - 5p mimics by hiperfect transfection reagents as experimen-tal group compared to negative control group(NC group)and blank group. Cell proliferation ability was evaluated by MTT assay. Cell migration ability was detected by transwell migration assay and wound healing assay. Cell invasion a-bility was detected by transwell invasion assay. Cell cycle and apoptosis were detected by flow cytometry. Results:There were no significant differences between mimics group and control groups by MTT assay(P ﹥ 0. 05). Transfec-tion of miR - 450a - 5p mimics into SKOV3 cells led to significant inhibition effect on cell migration and invasion a-bility detected by transwell assay(P ﹤ 0. 05). The migration ability of mimics group was significantly inhibited(P ﹤0. 05). Flow cytometry analysis displayed that miR - 450 a - 5 p significantly induced SKOV3 cell G1 arrest(P﹤ 0. 05),but no significant differences were found in cell apoptosis(P ﹥ 0. 05). Conclusion:miR - 450a - 5p over-expression may negatively regulate the invasion and migration ability of SKOV3 cells and may induce SKOV3 cells G1 arrest.%目的:研究 miR -450a -5p 对体外培养的人浆液性卵巢癌 SKOV3细胞增殖、迁移、侵袭、周期、凋亡机制的影响,分析 miR -450a -5p 对人卵巢癌 SKOV3细胞肿瘤生物学行为的影响。方法:运用 Hiperfect 转染试剂介导的转染法将 miR -450a -5p mimics 转染人卵巢癌 SKOV3细胞,设阴性对照组(NC 组)及空白对照组(blank 组)。采用 MTT 法、Transwell 迁移及侵袭实验、划痕实验、流式细胞术,分别检测细胞增殖、迁移及侵袭能力、周期及凋亡。结果:MTT 实验:miR -450a -5p mimics 组细胞增殖活性与 NC 组、blank 组无明显差异(P ﹥0.05)。Transwell 迁移及侵袭实验均显示转染 miR -450a -5p mimics 后,细胞的迁移及侵袭能力明显减弱(P ﹤0.05)。划痕后 miR -450a -5p mimics 组细胞迁移能力较 NC 组、blank 组降低(P ﹤0.05)。流式细胞术:转染 miR -450a -5p mimics 后,mimics 组细胞周期明显被阻滞于 G1期(P ﹤0.05)。miR -450a -5p mim-ics 组、NC 组、blank 组凋亡结果无明显差异(P ﹥0.05)。结论:过表达 miR -450a -5p 可能对人卵巢癌细胞系 SKOV3的侵袭及迁移能力存在负性调控,可能将 SKOV3细胞周期阻滞于 G1期。
