Slug 和 E-cadherin 在肾透明细胞癌组织中的表达及临床意义

摘要

Objective:To investigate the expression of Slug and E - cadherin in clear cell renal cell carcinoma (ccRCC)and its clinical significance. Methods:To detect the protein expression of Slug and E - cadherin in 83 ca-ses of ccRCC tissues and 40 cases of tumor - adjacent tissues by immunohistochemistry,detect the mRNA expression of Slug and E - cadherin in 20 cases of ccRCC and tumor - adjacent fresh tissues by Real - time PCR,analyze the relationship between Slug and E - cadherin protein and mRNA expression and the relationship between the protein expression of Slug,E - cadherin and clinicopathological parameters and prognosis. Results:The expression of Slug protein and mRNA in ccRCC tissue were significantly higher than that in the tumor - adjacent tissues,while the expression of E - cadherin protein and mRNA in ccRCC tissue were significantly lower than that in the tumor - adjacent tissues ( P <0. 05). There was significant negative correlation between Slug and E - cadherin protein and mRNA expression in ccRCC tissue( P < 0. 05). The expression of Slug protein was closely related with tumor size,TNM stages, lymph node metastasis,vascular invasion and recurrence( P < 0. 05). The expression of E - cadherin protein was closely related with gender,tumor size,TNM stages,lymph node metastasis and vascular invasion( P < 0. 05). The prognosis of patients with high expression of Slug protein was significantly worse than the patients with low expression (P <0. 05),while the prognosis of patients with low expression of E - cadherin protein was significantly worse than the patients with high expression of E - cadherin(P < 0. 05). Multivariate COX regression analysis showed that Slug and E - cadherin protein expression were the independent prognostic factors for patients with ccRCC. Conclusion: Slug and E - cadherin play an important role in the development of ccRCC,which can be used as biomarkers in judging the prognosis of ccRCC.%目的：研究 Slug 和 E - cadherin 在肾透明细胞癌（clear cell renal cell carcinoma，ccRCC）组织中的表达及其临床意义。方法：采用免疫组化 SP 法检测83例 ccRCC 组织和40例癌旁组织中 Slug 和 E - cadherin 蛋白的表达。采用 Real - time PCR 检测20例 ccRCC 及癌旁新鲜组织中 Slug 和 E - cadherin mRNA 的表达。分析二者蛋白和 mRNA 表达的相关性及 Slug 和 E - cadherin 蛋白表达与临床病理参数及预后的相关性。结果：Slug 蛋白和 mRNA 在 ccRCC 组织中的表达明显高于癌旁组织。E - cadherin 蛋白和 mRNA 在 ccRCC 组织中的表达明显低于癌旁组织（P ＜0.05）。ccRCC 组织中 Slug 和 E - cadherin 蛋白和 mRNA 表达均具有显著负相关性（P ＜0.05）。Slug 蛋白的表达与肿瘤大小、复发、TNM 分期、淋巴结转移及血管侵袭密切相关（P ＜0.05）；E - cadherin 蛋白的表达与性别、肿瘤大小、TNM 分期、淋巴结转移及血管侵袭密切相关（P ＜0.05）。Slug 蛋白高表达的患者预后明显差于低表达的患者（P ＜0.05）；而 E - cadherin 蛋白高表达的患者预后明显优于低表达的患者（P ＜0.05）。COX 多因素回归分析显示 Slug 和 E - cadherin 蛋白表达状态是影响 ccRCC患者预后的独立因素。结论：Slug 和 E - cadherin 在 ccRCC 发生发展中发挥重要作用，可作为 ccRCC 预后判断的生物标志物。
