单纯 LIM 蛋白在实体瘤中的研究进展

         

摘要

LIM - domain - only proteins(LMO)are a kind of containing zinc finger structure proteins,the proteins consist of four family members,including LMO1,LMO2,LMO3 and LMO4. Four members on the structure of the func-tional areas contain zinc finger structures,so they have similarities. Existing researches show that these proteins are not directly involved in the occurrence and development of tumor,but through to the related protein composition of protein complex interfere transcription and replication of chromosomes,thus tumor cell proliferation,invasion and metastasis are effected. These proteins earliest researches in malignant tumors were carried in T and B lymphocyte leukemia. The treatment about acute T lymphocyte leukemia with LMO2 gene mutations were carried out early. Researches about these proteins are active in the blood diseases. In recent years,researches about these proteins in neuroblastoma,glio-mas and diffuse large B cell lymphoma also gradually increased. There are many research reports in pancreatic cancer, prostate cancer and lung cancer. However,specific mechanism of action of these proteins still need further research, such as in what composition of protein complex effects in different tumors and tumor occurrence and development of complex proteins in their respective mechanism.%单纯 LIM 蛋白(LMO)是一类含有锌指结构的蛋白的总称,这类蛋白包含4个家族成员,分别为:LMO1、LMO2、LMO3、LMO4,这4个蛋白成员在功能区都有锌指结构,因此在结构上具有相似性。研究表明 LMO 并非直接参与肿瘤的发生和发展,而是通过与相关蛋白组成蛋白复合体影响肿瘤细胞增殖、侵袭和转移。LMO 在恶性肿瘤中的研究最早在急性 T 淋巴细胞白血病和急性 B 淋巴细胞白血病中展开,并且针对LMO2基因突变导致的急性 T 淋巴细胞白血病开展了早期的基因治疗。近年来 LMO 在神经母细胞瘤、神经胶质细胞瘤、弥漫大 B 细胞淋巴瘤、胰腺癌、前列腺癌以及肺癌等实体瘤细胞的研究也有不少报道。然而,这类蛋白的具体作用机制,如在不同肿瘤中与什么蛋白组成复合体影响肿瘤的发生、发展,复合体中蛋白的各自作用机理等问题还需更深入的研究。
